Abstract Optogenetic manipulation of neuronal activity through excitatory and inhibitory opsins has become an indispensable experimental strategy in neuroscience research. For many applications bidirectional control allowing both excitation inhibition the same neurons a single experiment is desired. This requires low spectral overlap between opsin, matched photocurrent amplitudes fixed expression ratio. Moreover, independent activation two distinct populations with different optogenetic actuators still challenging due to blue-light sensitivity all opsins. Here we report BiPOLES, tool for potent light wavelengths. BiPOLES enables sensitive, reliable dual-color spiking silencing single- or two-photon excitation, optical tuning membrane voltage, using second, sensitive opsin. The utility demonstrated worms, flies, mice ferrets.

Abstract In humans, traumatic social experiences can contribute to psychiatric disorders 1 . It is suggested that trauma impairs brain reward function such behaviour no longer rewarding, leading severe avoidance 2,3 rodents, the chronic defeat stress (CSDS) model has been used understand neurobiology underlying susceptibility versus resilience following trauma, yet little known regarding its impact on 4,5 Here we show that, CSDS, a subset of male and female mice, termed susceptible (SUS), avoid interaction with non-aggressive, same-sex juvenile C57BL/6J mice do not develop context-dependent encounters them. Non-social stressors have effect in either sex. Next, using whole-brain Fos mapping, vivo Ca 2+ imaging whole-cell recordings, identified population stress/threat-responsive lateral septum neurotensin (NT LS ) neurons are activated by interactions only SUS but resilient or unstressed control mice. Optogenetic chemogenetic manipulation NT their downstream connections modulates reward. Together, these data suggest previously rewarding targets possibly perceived as threats resulting from hyperactive occlude processing.

Fiber photometry is a key technique for characterizing brain-behavior relationships in vivo. Initially, it was primarily used to report calcium dynamics as proxy neural activity via genetically encoded indicators. This generated new insights into brain functions including movement, memory, and motivation at the level of defined circuits cell types. Recently, opportunity discovery with fiber has exploded development an extensive range fluorescent sensors biomolecules neuromodulators peptides that were previously inaccessible critical advance, combined availability affordable "plug-and-play" recording systems, made monitoring molecules high spatiotemporal precision during behavior highly accessible. However, while opening exciting avenues research, rapid expansion applications occurred without coordination or consensus on best practices. Here, we provide comprehensive guide help end-users execute, analyze, suitably interpret studies.

Male sexual behavior is innate and rewarding. Despite its centrality to reproduction, a molecularly specified neural circuit governing male reward remains be characterized. We have discovered developmentally wired necessary sufficient for mating. This connects chemosensory input BNSTprTac1 neurons, which innervate POATacr1 neurons that project centers regulating motor output reward. Epistasis studies demonstrate are upstream of BNSTprTac1-released substance P following mate recognition potentiates activation through Tacr1 initiate Experimental triggers mating, even in sexually satiated males, it rewarding, eliciting dopamine release self-stimulation these cells. Perception of threats is essential for survival. Previous findings suggest that parallel pathways independently relay innate threat signals from different sensory modalities to multiple brain areas, such as the midbrain and hypothalamus, immediate avoidance. Yet little known about whether how multi-sensory cues are integrated conveyed each modality amygdala, a critical area perception learning. Here, we report neurons expressing calcitonin gene-related peptide (CGRP) in parvocellular subparafascicular nucleus thalamus external lateral parabrachial brainstem respond various negative valence central respectively. Both CGRP populations their amygdala projections required aversive memory formation. The identification unified may provide insights into developing therapeutic candidates fear-related disorders.

Abstract The decision to either approach or avoid a potentially threatening environment is thought rely upon the coordinated activity of heterogeneous neural populations in hippocampus and prefrontal cortex (PFC). However, how this circuitry organized flexibly promote both avoidance at different times has remained elusive. Here, we show that hippocampal projection PFC composed two parallel circuits located superficial deep pyramidal layers CA1/subiculum border. These have unique upstream downstream connectivity, are differentially active during behaviour. population preferentially connected widespread inhibitory interneurons, its activation promotes exploration; while circuit neurons fast spiking avoidance. Together provides mechanism for regulation behaviour conflict: through specialized, allow bidirectional control PFC.
