The claustrum (CLA), a subcortical nucleus in mammals, essentially composed of excitatory projection neurons and known for its extensive connections with the neocortex, has recently been associated variety functions ranging from consciousness to impulse control. However, research on CLA challenging due difficulties specifically comprehensively targeting neuronal populations. In various cases, this limitation led inconsistent findings lack reliable data. present work, we describe expression profile Smim32 gene, which is almost exclusively transcribed endopiriform nucleus, as well inhibitory thalamic reticular nucleus. Persistent pain is a prevalent medical concern correlating with hyperexcitable anterior cingulate cortex (ACC). Its activity modulated by inputs from several brain regions, but the maladjustments that these afferent circuits undergo during transition acute to chronic still require clarification. We focus on ACC-projecting claustrum (CLAACC) neurons and their responses sensory aversive stimuli in mouse model of inflammatory pain. Using chemogenetics, vivo calcium imaging, ex electrophysiological approaches, we reveal suppression CLAACC acutely attenuates allodynia preferentially transmits information ACC. With prolonged pain, claustro-cingulate functional impairment develops, which mediated weakened excitatory drive onto ACC pyramidal neurons, resulting diminished claustral influence These findings support an instrumental role processing nociceptive its susceptibility persistent states.

Abstract The claustrum has been linked to attention and sleep. We hypothesized that this reflects a shared function, determining responsiveness stimuli, which spans the axis of engagement. To test hypothesis, we recorded population dynamics from male mice during both sleep an attentional task (‘ENGAGE’). Heightened activity in neurons projecting anterior cingulate cortex (ACCp) corresponded reduced sensory Similarly, ENGAGE task, heightened ACCp correlated with disengagement behavioral lapses, while low hyper-engagement impulsive errors. Chemogenetic elevation awakenings errors task. Furthermore, employing exploration strategy showed stronger correlation between performance compared exploitation complexity. The claustrum is thought to be one of the most highly interconnected forebrain structures but its organizing principles have yet fully explored at level single neurons. Here, we investigated identity, connectivity, and activity identified neurons understand how structure's unique convergence input divergence output support binding information streams. Neural activity in the claustrum has been associated with a range of vigilance states, yet patterns and efficacy synaptic communication identified neurons have not thoroughly determined. Here, we show that projecting to retrosplenial cortex are most active during synchronized cortical states such as non-rapid eye movement (NREM) sleep suppressed increased desynchronization arousal, movement, REM sleep. The claustrocortical signaling is NREM diminished due part cholinergic tone. Finally, activation enhances memory consolidation through phase resetting delta waves. Therefore, constrained function effectively cognitive processes consolidation.
