Abstract Genetically encoded calcium (Ca 2+ ) indicators (GECIs) are widely used for imaging neuronal activity, yet current limitations of existing red fluorescent GECIs have constrained their applicability. The inherently dim fluorescence and low signal-to-noise ratio red-shifted posed significant challenges. More critically, several red-fluorescent exhibit photoswitching when exposed to blue light, thereby limiting applicability in all-optical experimental approaches. Here, we present the development PinkyCaMP, first mScarlet-based Ca sensor that outperforms sensors brightness, photostability, ratio, compatibility with optogenetics neurotransmitter imaging. PinkyCaMP is well-tolerated by neurons, showing no toxicity or aggregation, both vitro vivo . All approaches, including single-photon excitation methods such as fiber photometry, widefield imaging, miniscope well two-photon awake mice, fully compatible PinkyCaMP.

The ability to evaluate valence of a social agent based on experience is essential for an animal's survival in its group1. Although hippocampal circuits have been implicated distinguishing novel and familiar conspecifics2-7, it remains unclear how constructed the basis history what mechanisms underlie heightened versatility dynamic relationships. Parkinson's disease (PD) is characterized by early-onset olfactory dysfunction preceding motor symptoms, yet its mechanisms remain elusive. Based on the studies microbiota-gut-brain axis, microbiota-nose-brain axis might be involved in pathogenesis of PD. However relative are rare. By consecutive 14-days intranasally transplanting bacteria, we established mice models exhibiting nasal microbiota dysbiosis (NMD), including animal group received intranasal drops fecal bacterial suspension from normal (NB group) and PD (PB group), with animals that only anesthesia used as control group. Then analyzed composition via 16S rRNA sequencing, evaluated functions through behavioral experiments, buried food test, open field pole descent traction test.
