Objective This research aims to investigate putative mechanisms between glymphatic activity and cognition in mild cognitive impairment (MCI) analyzes whether the relationship reserve (CR) was mediated by activity. Methods 54 MCI patients 31 NCs were enrolled evaluate bilateral diffusivity along perivascular spaces acquire an index for space (ALPS-index) on diffusion tensor imaging (DTI). The year of education used as a proxy CR. ALPS-index compared two groups correlation analyses among ALPS-index, function, CR conducted. Mediation applied correlations CR, cognition. Results group had significantly lower right whole brain but higher y-axis projection fiber area (Dyproj) than NCs. In group, left Dyproj negatively related test scores positively correlated with analysis demonstrated that partially function. Conclusion exhibited decreased has protective effect against decline MCI, this may be changes

More than 5 years have passed since the Diffusion Tensor Image Analysis ALong Perivascular Space (DTI-ALPS) method was proposed with intention of evaluating glymphatic system. This is handy due to its noninvasiveness, provision a simple index in straightforward formula, and possibility retrospective analysis. Therefore, ALPS adopted evaluate system for many disorders studies. The purpose this review look back discuss at moment.

Abstract Recently, an astrocytic aquaporin 4-dependent drainage system, that is, the glymphatic has been identified in live murine and human brain. Growing evidence suggests function is impaired patients with several neurodegenerative diseases, including Alzheimer’s Parkinson’s disease. As third most common disease, although animal studies have indicated early dysfunction likely important pathological mechanism underpinning amyotrophic lateral sclerosis (ALS), no available study conducted to thoroughly assess vivo ALS date, particularly early-stage ALS. Thus, using diffusion tensor imaging analysis along perivascular space (ALPS) index, approximate measure of vivo, we aimed explore whether ALS, diagnostic performance ALPS index distinguishing between healthy subjects. We also identify relationships clinical disabilities sleep problems In this retrospective study, King’s Stage 1 were defined as enrolled 56 32 age- sex-matched control All participants completed screening, assessment analysis. For assessment, Pittsburgh Sleep Quality Index, Epworth Sleepiness Scale polysomnography used. Compared subjects, had a significantly lower after family-wise error correction (P < 0.05). Moreover, receiver operating characteristic showed area under curve for was 0.792 (95% confidence interval 0.700–0.884). Partial correlation analyses correlated disability disturbances Multivariate efficiency (r = 0.419, P 0.002) periodic limb movements −0.294, 0.017) significant predictive factors conclusion, our continues support role pathology, provide additional insights into value its suggest improvement may be promising strategy slowing process Future are needed therapeutic individuals presymptomatic-stage diseases.

Abstract Cerebrospinal fluid (CSF) plays a critical role in metabolic waste clearance from the brain, requiring its circulation throughout various brain pathways, including ventricular system, subarachnoid spaces, para‐arterial interstitial and para‐venous spaces. The complexity of CSF has posed challenge obtaining noninvasive measurements dynamics. assessment dynamics circulatory pathways is possible using diffusion magnetic resonance imaging (MRI) with optimized sensitivity to incoherent water movement across brain. This review presents an overview both established emerging MRI techniques designed measure their potential clinical applications. discussion offers insights into optimization acquisition parameters enhance specificity metrics on underlying Lastly, we emphasize importance cautious interpretations diffusion‐based imaging, especially when differentiating between tissue‐ fluid‐related changes or elucidating structural versus functional alterations.

Abstract Purpose To investigate glymphatic system function in patients with brain tumors, including both primary and secondary using diffusion tensor imaging along perivascular spaces (DTI-ALPS). Methods We retrospectively analyzed the MR DTI of 24 unilateral tumors compared them age sex-matched controls. DTI-ALPS index ipsi- contralateral hemispheres. The region interest was placed periventricular vessels adjacent to lateral ventricles. Differences between sex, age, kind tumor (primary or metastasis) were evaluated. Correlations tumor's apparent coefficient (ADC) also investigated. Results significantly lower ( p < 0.05) tumor-affected hemisphere (mean = 1.26 ± 0.24) than 1.43 0.28). A comparison healthy controls revealed no significant difference on matched ipsilateral side. However, side larger HC. Additionally, statistically differences found when analyzing vs. entity. we did not find a correlation patient ADC. Conclusion decreased may be related impaired function. cancer is often systemic disease; thus, from generally considered as normal control. Nonetheless, does only reflect but it can influenced by factors such axonal degeneration. Therefore, directly waste clearance changes should interpreted carefully.

The recently discovered glymphatic system may support the removal of neurotoxic proteins, mainly during sleep, that are associated with neurodegenerative diseases such as Alzheimer's and Parkinson's Disease. Diffusion tensor image analysis along perivascular space (DTI-ALPS) has been suggested a method to index health (with higher values indicating more intact system). Indeed, in small-scale studies DTI-ALPS shown correlate age, cognitive health, is females than males. To determine whether these relationships stable we replicated previous findings associating demographic, sleep-related, markers large sample participants from UK Biobank. We calculated Biobank (n = 17723). Using Bayesian Frequentist approaches, replicate previously reported between index. found predicted associations longest uninterrupted sleep window (LUSWT) on typical night, performance, sex. However, effects were substantially smaller those studies. Parameter estimates this study be used priors subsequent using approach. These results suggest consistently, therefore predictably, demographics, LUWST, cognition. propose metric, for first time large-scale, population-based cohort, measure, but one which stronger links function needed before it can understand outcomes UKBiobank.
