Bupleurum chinense DC.-Scutellaria baicalensis Georgi (BS) is a classic drug pair that has good clinical effects on depression and many tumors. Abstract Major depressive disorder (MDD) is a severe mental associated with high rates of morbidity and mortality. Current first‐line pharmacotherapies for MDD are based on enhancement monoaminergic neurotransmission, but these antidepressants still insufficient produce significant side‐effects. Consequently, the development novel therapeutic targets desired. Engeletin, natural Smilax glabra rhizomilax derivative, compound proven efficacy in treating ischemic stroke, yet its effects mechanisms depression remain unexplored. The engeletin were assessed forced swimming test (FST) tail suspension (TST) mice. Engeletin was also investigated chronic restraint stress (CRS) mouse model fluoxetine (FLX) as positive control. Changes prefrontal cortex (PFC) spine density, synaptic plasticity‐linked protein expressions brain‐derived neurotrophic factor (BDNF)‐tyrosine kinase B (TrkB)‐ mammalian target rapamycin complex 1 (mTORC1) signalling pathway after treatment then investigated. TrkB mTORC1 selective inhibitors, ANA‐12 rapamycin, respectively, utilized to assess engeletin's antidepressive mechanisms. Our data shows that exhibited antidepressant‐like activity FST TST mice without affecting locomotor activity. Furthermore, it efficiency against CRS model. The prevention, mitigation and treatment of depression has become a global issue that needs to be solved urgently. Sayram Ketteki, traditional natural fermented yoghurt from the region with world's fourth highest life expectancy, been known as "longevity secret", whose longevity anti-depression factors are speculated come its rich microorganisms. Therefore, for first time, we systematically studied in depth microbes screened new edible probiotic strain, Lactiplantibacillus plantarum R6-3, explored effect chronic unpredictable mild stress (CUMS)-induced mice. It is encouraging L. R6-3 was significantly superior classic anti-depressant drug, fluoxetine, performance promoting sucrose preference test (SPT) behavior by 18% (p < 0.001), lowering serum CORT content 5.6% 0.05), accelerating brain-derived neurotrophic factor (BDNF) level 5.9% 0.01), increasing IL-10 concentration 2.3% up-regulating expression BDNF phosphorylated-ERK 74% 0.01) 45% respectively, facilitating secretion fecal short-chain fatty acids (SCFAs), including n-butyric, n-valeric, isovaleric acid 47% 42% 0.05) 38% respectively.
