Tumor-associated macrophages (TAMs) are present in almost all solid tumor tissues. 16They play critical roles immune regulation, angiogenesis, stem cell activation, invasion and metastasis, resistance to therapy. However, it is unclear how TAMs perform these functions. With the application of single-cell RNA sequencing (scRNA-seq), has become possible identify TAM subpopulations associated with distinct In this review, we discuss four novel tumors based on core gene signatures by scRNA-seq, including FCN1 + , SPP1 C1Q CCL18 TAMs. Functional enrichment expression scRNA-seq data from different tissues found that may induce inflammation; potentially involved cancer whereas participate regulation suppression; And cells terminal immunosuppressive not only have a stronger function but also enhance metastasis. can be further divided into populations Meanwhile, will emerging evidence highlighting separating macrophage there exist potential disconnects between types identified their actual function.

Despite the acknowledged advantages of combined immunochemotherapy for tumor treatment, high efficiency co-delivery these agents into targeted tissue is still challenging. Herein, based on a "three-birds-with-one-stone" strategy, facile glycyrrhizic acid (GL)-lipid hybrid nanoplatform loading triptolide (TP/GLLNP) designed to better address dilemma. Differing from traditional liposomes with dual-drug NPs, GL cholesterol-like structure primarily employed construct lipid membrane skeleton GL-based nanoparticle (GLLNP), and then (TP) readily loaded in bilayer GLLNP. The fabricated GLLNP possessed similar drug efficacy, particle size, storage stability; none hemolysis; even higher fluidity; lower absorbed opsonin proteins compared conventional liposomes. Compared TP-loaded (TP/Lipo), TP/GLLNP exhibits significantly enhanced cellular uptake, cytotoxicity, apoptosis HepG2 cells. In addition, could ameliorate immunosuppression by promoting tumor-associated macrophage polarization M2 M1 phenotype. Furthermore, retention accumulation area be found. As expected, displayed synergistic anti-hepatocellular carcinoma efficacy vivo. conclusion, this study provides an inspirational strategy combine anti-HCC benefits TP using novel nanosystem.

Abstract Background Histone lysine lactylation ( Kla ) is a newly identified histone modification, which plays crucial role in cancer progression. Hence, we determined the prognostic value of breast (BC). Methods We obtained RNA expression profiles BC from The Cancer Genome Atlas (TCGA), following screening out Kla-specific genes. Furthermore, by constructing cox model based on Subsequently, lactate accumulation-related genes and through Human Protein HPA ), further performed correlation analysis their expression. Meanwhile, explored effects tumor microenvironment (TME), drug therapy immunotherapy. Moreover, predicted pathways influenced via gene set enrichment (GSEA). Results A total 1073 samples 112 normal controls were TCGA, 23 removed owing to inadequate clinical information. 257 differentially expressed DEKlaG s) BC. involved with CCR7 , IGFBP6 NDUFAF6 OVOL1 SDC1 was established, risk score could be visualized as an independent biomarker for remarkably associated immune microenvironment, related activation various BC-related KEGG pathways. Conclusion In conclusion, contributes resistance undesirable responses, prognosis, suggesting that expected new therapeutic target

Purpose: Chronic gastroesophageal reflux disease (GERD) causes the abnormal of acid and bile salts, which would induce Barrett's esophagus (BE) esophageal adenocarcinoma (EAC). EGFR, as one main components exosome, plays an important role in cancer progression. Here, we investigated acidic salts (ABS)-induced exosomal EGFR EAC cell proliferation. Methods: Electronic microscopic examination Western blot were used to identify exosomes. blot, siRNA transfection, enzyme-linked immunosorbent assay, qRT-PCR, viability detection, mouse xenograft tumor models, immunohistochemical staining performed study function ABS-induced Results: We found that ABS improved level normal human epithelial cells, BE BE-associated cells. The results confirmed serum-derived exosomes from healthy persons patients suffering GERD, with or without GERD. Moreover, line-derived was promote macrophage M2 polarization through PI3K-AKT pathway. co-incubation medium macrophages proliferation growth, depended on level. CCL18 identified most effective component by binding its receptor PITPNM3 vitro vivo. Conclusion: Our findings demonstrate regulates This provides insight into development. AGS-30, a new andrographolide derivative, showed significant anticancer and anti-angiogenic characteristics. However, its role in controlling macrophage polarization tumor immune response is unknown. Thus, the main goals of this study are to investigate how AGS-30 regulates it suppresses breast cancer metastasis. inhibited IL-4 IL-13-induced RAW 264.7 THP-1 macrophages into M2-like phenotype. did not affect LPS IFN-γ-induced M1-like macrophages. reduced mRNA expressions CD206, Arg-1, Fizz-1, Ym-1, VEGF, IL-10, MMP2, MMP9 concentration-dependent manner. In contrast, treatment at 5 μM polarization. The conditioned medium from increased 4T1 cell migration invasion, whereas these effects. xenograft mice, volume weight were without affecting body after receiving AGS-30. also lung liver metastasis, with STAT6, CD31, Ki67 protein expressions. Moreover, tumors had considerably fewer Arg-1 expression, but proportion iNOS expression treatment. Same results found tail vein metastasis model. conclusion, shows that inhibits growth probably through inhibiting Our findings suggest may be potential immunotherapeutic alternative for metastatic cancer.
