Scope Previous study has demonstrated the chemical structure of chondroitin sulfate (CHS) from Halaelurus burgeri skin and its effects on insulin resistance. However, precise impact this phenomenon endoplasmic reticulum (ER) stress inflammation, which contribute to resistance, remains unclear. This is investigate CHS ER stress, inflammatory response signaling, gut microbiota in high‐fat diet (HFD)‐fed mice. Methods results HFD‐fed C57BL/6J mice receive dietary gavage intervention for 18 weeks. Blood, liver tissue, feces are harvested further investigation. Results show that inhibits accompanied by lowered blood glucose, nitric oxide (NO), reactive oxygen (ROS), free fatty acids (FFA) levels, increases hepatic glycogen accumulation. Moreover, inflammation improved treatment via inactivation Toll‐like receptor 4 (TLR4) signaling downstream c‐jun N‐terminal kinase (JNK) nuclear factor kappa B (NFκB) pathways. Additionally, regulates microbiota, particularly decline Firmicutes Bacteroidetes ratio. also lowers fecal lipopolysaccharide elevates several short chain acids. Conclusion These findings suggest H. may be an alternative functional food supplement anti‐ER anti‐inflammtion, regulation microbiota.

Acute kidney injury (AKI) represents significant medical challenges due to its elevated rates of morbidity and mortality, with limited therapeutic options currently available. Hence, the exploration novel medicinal treatments for AKI management remains vital. Substances medicine food homology (SMFH), referring substances having characteristics both medicine, have been applied in China thousands years. They could be used daily diets body conditioning. Traditional Chinese (TCM), naturally derived components demonstrated effectiveness, presents distinctive benefits treatment. Numerous studies shown that SMFH TCM phytochemicals function satisfactorily nephroprotective effects a effect on alleviating as well complications. In this review, pathogenesis was illustrated. We concentrated against tried summarize underlying mechanisms various kinds AKI, highlighting crucial phytochemical prevention therapy. Besides, strategies globalization are analysed. This review comprehensively reveals exhibit promising potential intervention by targeting signal pathways targets, which would contribute AKI's cognition, preventive treatments, global promotion.

Acute kidney injury (AKI) remains a critical condition with limited therapeutic options, predominantly managed by renal replacement therapy. The challenge of developing targeted treatments persists. We integrated genetic data related to druggable proteins and gene expression AKI genome-wide association study (GWAS) findings. Based on multi-omics Mendelian randomization (MR), we identified the potential causal influence 5,883 unique genes AKI. also performed using reverse MR external cohort-based analysis verify robustness this relationship. Expression patterns these targets were examined bulk transcriptome single-cell data. In addition, drug repurposing analyses conducted explore existing medications. constructed molecular interaction network interplay between known drugs. Genetically predicted levels seven twelve associated an increased risk Of these, six (NCF1, TNFRSF1B, APEH, ACADSB, ADD1, FAM3B) prioritized based robust evidence validated in independent cohorts. Reverse showed one-way relationship targets. These are expressed proximal tubular cells, endothelial collecting duct-principal immune cells within both AKI-affected normal tissues. Several promising opportunities identified, such as telmisartan-NCF1, calcitriol-ACADSB, ethinyl estradiol-ACADSB. mapping pathway integration provided further insights, suggesting strategies for combinatorial therapies. This extensive investigation several highlighted repurposing. findings offer valuable insights that could shape future research development treatments.

The aims of this study were to explore the protective effect Xiaochaihu decoction in mice with sepsis induced by intraperitoneal injection; its anti-inflammatory on TLR4-MyD88-NF-κB signalling pathway; and main material basis decoction, aim supplementing expanding associated research providing a scientific foundation for clinical use decoction. effects septic analysed measurements white blood cells (WBC) Platelets (PLT); Nitric Oxide (NO) level serum; IL-6, IL-1β TNF-α levels RT-PCR; Haematoxylin-Eosin (HE) immunohistochemistry; western blotting (WB). results showed excellent vivo LPS-induced mice, through down regulation gene protein expression TLR4, MYD88, TRAF6, IKK, IKBα p65 subsequent reduction release inflammatory mediators IL-1β, NO. Moreover, significant anti-septic was observed from high medium doses but not low dose.
