Nanoplastics (NPs) are ubiquitous in the environment, posing significant threats to biological systems, including nervous across various trophic levels. Nevertheless, molecular mechanisms behind size-dependent neurotoxicity of NPs remain unclear. Here, we investigated 20 and 100 nm polystyrene (PS-NPs) zebrafish. Utilizing dynamics simulations complementary methods, discovered that PS-NPs initiated by promoting dimerization toll-like receptor 4/myeloid differentiation-2 (TLR4/MD-2) complex. This process involves binding hydrophobic pocket MD-2, which induced flipping Phe-126 toward dimer interface bending C-terminal domain TLR-4, bringing two domains into close proximity. Thereafter, astrocytes microglia were activated, initiating a cascade events include neuroinflammation, central system cell apoptosis, inhibition motor neuron development, ultimately alteration swimming behavior Further, elicited more severe than PS-NPs, attributed their higher accumulation brain as determined through 14C-labeled effective interaction with TLR4/MD-2 Overall, our study uncovers underlying NPs, merit attention during risk assessment regulation.
