Bis(2-ethylhexyl) phthalate (DEHP) is an endocrine disruptor that may cause damage to several species. Quercetin (Que), a common flavonoid, has anti-inflammation, antioxidation, and immune regulation properties. In this study, we identified DEHP-exposed or Que-antagonist groups in chicken MSB-1 cells explore whether Que can mitigate DEHP-caused bursa of Fabricius pyroptosis necroptosis. The findings demonstrated reduced the expression necroptosis pyroptosis, inhibited mitogen-activated protein kinase (MAPK)/NF-κB pathway, mitigated oxidative stress caused by DEHP. addition reactive oxygen species activator (Sanguinarin) raised extent stress, NF-κB (nuclear factor-kappa-B activator1, act1) activated MAPK/NF-κB pathway compared + DEHP group. conclusion, counteract DEHP-induced programmed necrosis. In recent years, the study of microplastics (MPs) and nanoplastics (NPs) their effects on human health has gained significant attention. The impacts NPs lipid metabolism specific mechanisms involved remain poorly understood. To address this, we utilized high-throughput sequencing molecular biology techniques to investigate how endoplasmic reticulum (ER) stress might affect hepatic in presence polystyrene (PS-NPs). Our findings suggest that PS-NPs activate PERK-ATF4 signaling pathway, which turn upregulates expression genes related synthesis via ATF4-PPARγ/SREBP-1 pathway. This activation leads an abnormal accumulation droplets liver. 4-PBA, a known ER inhibitor, was found mitigate PS-NPs-induced disorder. Nanoplastics (NPs) are emerging contaminants having persistent nature, diverse ecological impacts, and potential food safety risks. Here, we examined the ecotoxicity of 80 nm polystyrene nanoplastics (PS-NPs) at environmentally relevant concentrations (ERCs, 10 100 μg/L), sublethal (SLCs, 500 2500 μg/L) in Magallana hongkongensis. Results showed that SLCs significantly (p < 0.05) increased superoxide dismutase (SOD), catalase (CAT), alkaline phosphatase (AKP) activities altered tnfα, cat, gst, sod, se-gpx genetic expressions. Further, PS-NP exposure both levels reduced beneficial bacteria potentially pathogenic gut. In transcriptomic analysis, 5118 4180 differentially expressed genes (DEGs) were identified ERCs, while 5665 4817 DEGs found SLCs, respectively. Upregulated enriched lysosomes, ABC transporters, apoptosis pathways, downregulated ribosomal pathways. Overall, ERCs gut microbiota responses, addition, also impacted antioxidant immune systems.

As emerging environmental contaminants, nanoplastics (NPs) are progressively accumulating in terrestrial and aquatic ecosystems worldwide, posing a potential threat to human health. The liver is considered as one of the primary organs targeted by NPs accumulation living organisms. However, there remains large knowledge gap concerning NPs-induced hepatotoxicity. In this study, we examined impact chronic exposure environmentally relevant doses polystyrene (PS) on hepatic pyroptosis mice. results demonstrated that both particle sizes PS-NPs (100 nm 500 nm) significantly triggered mouse liver, evidenced upregulation GSDMD-N protein levels; moreover, pyroptotic effect induced 100 was more pronounced compared PS-NPs. Mechanistically, resulted an TXNIP expression, thereby activating NLRP3 inflammasome subsequently inducing inflammatory responses pyroptosis. Notably, following termination subsequent recovery period 50 days, PS-NPs-mediated inflammation via TXNIP/NLRP3 pathway were effectively ameliorated, even returning levels close baseline. Collectively, our findings provide novel evidence for size-dependence reversibility through TXNIP/NLRP3/GSDMD vivo.
