Abstract Gastrointestinal cancers, a major global cause of cancer‐related mortality and disease burden, are heterogeneous group malignant aliments involving different organs the digestive system. The late clinical diagnosis, genomic tumor heterogeneity, high complexity gastrointestinal microenvironment, along with increasing treatment resistance have been recognized as main contributing factors to current inadequacy interventions poor prognosis cancer patients. Biomembrane‐coated biomimetic nanoparticles (NPs) recently appointed advanced nanotechnological tools for management cancers. Abstract The study presents a transcriptomics‐based liquid biopsy approach for early recurrence detection in locally advanced gastric cancer (LAGC). Four mRNA biomarkers (AGTR1, DNER, EPHA7, and SUSD5) linked to are identified through transcriptomic data analysis. A Risk Stratification Assessment (RSA) model combining these with clinical features showed superior predictive accuracy postoperative recurrence, AUCs of 0.919 0.935 surgical validation cohorts, respectively. Functional studies using human cell lines AGS HGC‐27 demonstrated that silencing the panel genes impaired migration, invasion, proliferation. In vivo experiments further reduced tumor growth, metastasis, lymphangiogenesis mice, possibly mediated by cAMP signaling pathway. This non‐invasive offers significant potential enhancing enabling personalized treatment strategies, thereby improving patient outcomes management LAGC.
