Necrotizing enterocolitis (NEC) is a gastrointestinal complication of premature infants with high rates morbidity and mortality. A comprehensive view the cellular changes aberrant interactions that underlie NEC lacking. This study aimed at filling in this gap. We combine single-cell RNA sequencing (scRNAseq), T-cell receptor beta (TCRβ) analysis, bulk transcriptomics, imaging to characterize cell identities, interactions, zonal NEC. find an abundance proinflammatory macrophages, fibroblasts, endothelial cells as well T exhibit increased TCRβ clonal expansion. Our analyses highlight dysregulations NEC-associated intestinal tissue identify potential targets for biomarker discovery therapeutics.
