Hepatitis B virus (HBV) infection remains a major global health concern, necessitating the development of sensitive and reliable diagnostic methods. In this study, we propose novel approach to enhance sensitivity HBV DNA detection by leveraging concentration imbalance-driven circuit (CIDDC) as an operational amplifier, coupled with hybridization-responsive DNA-templated silver nanocluster (DNA-AgNCs) nanoprobe named Q·C6-AgNCs. The CIDDC system effectively converts amplifies input into enriched generic single-stranded output, which subsequently triggers fluorescence DNA-AgNCs reporter upon hybridization, generating measurable signal for detection. By incorporating circuit, not only achieved enhanced lower limit 0.11 nM but also demonstrated high specificity single-base mismatch discriminability Additionally, mix-and-detect assay format is simple, user-friendly, isothermal. This innovative strategy holds promise advancing molecular diagnostics facilitating effective management HBV-related diseases.

Berberine (BBR), as a natural isoquinoline alkaloid, has been demonstrated various pharmacological activities, and widely applied in the treatment of diseases. The quantitative analysis BBR is important for its researches clinical applications. In work, utilizing specific interaction between triplex DNA, sensitive selective fluorescent detecting method was established with DNA-templated silver nanoclusters (DNA-AgNCs). After binding structure template DNA-AgNCs, quenched fluorescence DNA-AgNCs formed BBR-triplex complex yellow-green fluorescence. ratiometric signal showed linear relationship concentration range 10 nM to 1000 nM, detect limit nM. Our exhibited excellent sensitivity selectivity, it further detection real samples.
