Novel coronavirus disease 2019 (COVID-19) has resulted in a global pandemic and is caused by severe acute respiratory syndrome 2 (SARS-CoV-2). Several studies have suggested that precise disulfide-thiol balance crucial for viral entry fusion into the host cell oxidative stress generated from free radicals can affect this balance. Here, we reviewed current knowledge about role of on SARS-CoV SARS-CoV-2 infections. We focused impact antioxidants, like NADPH glutathione, redox proteins, such as thioredoxin protein disulfide isomerase, maintain cell. The possible influence these biomolecules binding with angiotensin-converting enzyme II receptor well severity COVID-19 infection was discussed.

The disease severity of COVID-19, especially in the elderly and patients with co-morbidities, is characterized by hypercytokinemia, an exaggerated immune response associated uncontrolled excessive release proinflammatory cytokine mediators (cytokine storm). Flavonoids, important secondary metabolites plants, have long been studied as therapeutic interventions inflammatory diseases due to their cytokine-modulatory effects. In this review, we discuss potential role flavonoids modulation signaling pathways that are crucial for COVID-19 disease, particularly those related inflammation immunity. immunomodulatory ability flavonoids, carried out regulation mediators, inhibition endothelial activation, NLRP3 inflammasome, toll-like receptors (TLRs) or bromodomain containing protein 4 (BRD4), activation nuclear factor erythroid-derived 2-related 2 (Nrf2), might be beneficial regulating storm during SARS-CoV-2 infection. Moreover, inhibit dipeptidyl peptidase (DPP4), neutralize 3-chymotrypsin-like protease (3CLpro) affect gut microbiota maintain response, dual action angiotensin-converting enzyme (ACE-2) may potentially also applied responses induced SARS-CoV-2. Based on previously proven effects other basis newly published studies (bioinformatics, molecular docking), it reasonable assume positive changes COVID-19. This review highlights current state knowledge utility management points multiple biological processes deregulated pathology identification agents, including naturally occurring substances such represents great approach utilizable Although not clinically investigated yet, applicability against could a promising strategy broad spectrum activities.

Abstract The outbreak of COVID-19 has become a global crisis, and brought severe disruptions to societies economies. Until now, effective therapeutics against are in high demand. Along with our improved understanding the structure, function, pathogenic process SARS-CoV-2, many small molecules potential anti-COVID-19 effects have been developed. So far, several antiviral strategies were explored. Besides directly inhibition viral proteins such as RdRp M pro , interference host enzymes including ACE2 proteases, blocking relevant immunoregulatory pathways represented by JAK/STAT, BTK, NF-κB, NLRP3 pathways, regarded feasible drug development. development treat achieved strategies, computer-aided lead compound design screening, natural product discovery, repurposing, combination therapy. Several representative remdesivir paxlovid proved or authorized emergency use countries. And candidates entered clinical-trial stage. Nevertheless, due epidemiological features variability issues it is necessary continue exploring novel COVID-19. This review discusses current findings for treatment. Moreover, their detailed mechanism action, chemical structures, preclinical clinical efficacies discussed.

The adverse effects of Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) are not limited to the related infectious disease. In children and adolescents, serious risks due coronavirus disease 2019 (COVID-19) pandemic also its indirect effects. These include an unbalanced diet with increased risk weight excess or nutritional deficiencies, sedentary lifestyle, lack schooling, social isolation, impaired mental health.Pediatricians should be aware side COVID-19 on children's diet, physical health advise families according their needs financial resources. Moreover, a targeted therapy able offer protection against deleterious SARS-CoV-2 infection require greater effort by scientific societies find more effective prevention strategy. this context, much interest given support, contrast malnutrition stimulate immune system.
