The Chinese giant salamander (CGS), Andrias davidianus, the largest living amphibian, is of significant conservation importance for its wild populations and serves as a valuable economic species in aquaculture. Given that male CGS are larger than female, understanding mechanisms behind gonadal differentiation development critical optimizing production This study conducted small RNA-seq on female gonads during two key windows CGS. PCA analysis revealed distinct clustering within groups clear between groups. A total 374 miRNAs were identified, including 162 known 212 novel miRNAs. Differential expression enrichment across time points identified nine sex-biased involved differentiation, let-7b-3p, miR-3529-3p, miR-34c-5p, miR-10-5p, miR-7-5p, four miRNAs, network these with their target genes constructed. Abstract Triploids are rare in nature because of difficulties meiotic and gametogenic processes, especially vertebrates. The Carassius complex cyprinid teleosts contains sexual tetraploid crucian carp/goldfish ( C. auratus ) unisexual hexaploid gibel carp/Prussian carp gibelio lineages, providing a valuable model for studying the evolution maintenance mechanism polyploids Here we sequence genomes two species assemble their haplotypes, which contain subgenomes (A B), to chromosome level. Sequencing coverage analysis reveals that is an amphitriploid (AAABBB) with triploid sets chromosomes; each set derived from different ancestor. Resequencing data strains show reproduction has been maintained over 0.82 million years. Comparative genomics intensive expansion alterations cell cycle-related genes oocyte-specific histone variant. Cytological assays indicate produces unreduced oocytes by alternative ameiotic pathway; however, sporadic homologous recombination high rate gene conversion also exist . These genomic changes might have facilitated purging deleterious mutations maintaining genome stability this fish. Aquaculture originates from ancient Chinese wisdom, and is considered the most efficient food-producing industry. About 70 years ago, New China proposed an industrial approach to fish production made a policy decision begin modern aquaculture development. As one of lucky persons since early 1980s, I have committed my scholarly career studies on genetics breeding biotechnology polyploid gibel carp (Carassius gibelio). Here, several significant recent innovations, such as proposition amphitriploid concept, association ploidy changes with reproduction transition, finding unique ameio-fusiongenesis mode, establishment controllable fertility approach, creation intermuscular bone-free mutants, well recreation desirable goldfish, are introduced emphasized active participant these innovations.
