Adverse early-life experiences (ELA), including poverty, trauma and neglect, affect a majority of the world's children. Whereas impact ELA on cognitive emotional health throughout lifespan is well-established, it not clear how distinct types influence child development, there are no tools to predict for an individual their vulnerability or resilience consequences ELAs. Epigenetic markers DNA-methylation profiles peripheral cells may encode provide predictive outcome marker. However, rapid dynamic changes in DNA methylation childhood inter-individual variance human genome pose barriers identifying predicting outcomes exposure. Here, we examined relation several dimensions methylation, using longitudinal within-subject design high threshold hope mitigating above challenges.

Studies reporting accelerated ageing in children with affective disorders or maltreatment exposure have relied on algorithms for estimating epigenetic age derived from adult samples. These limited validity estimation during early development. We here use a pediatric buccal (PedBE) clock to predict DNA methylation-based deviation and without internalizing disorder assess the moderating effect of exposure. further conduct gene set enrichment analysis contribution glucocorticoid signaling PedBE clock-based results.DNA was isolated saliva 158 [73 girls, 85 boys; mean (SD) = 4.25 (0.8) years] including Epigenetic estimated based methylation across 94 CpGs clock. Adverse stressful experiences in utero can redirect fetal brain development, ultimately leading to increased risk for psychiatric disorders. Obesity during pregnancy have similar effects as maternal stress, affecting mental health the offspring. In order explain how outcomes may originate from different prenatal conditions, we propose a "funnel effect" model whereby psychological or metabolic stress triggers same evolutionarily conserved response pathways, increasing vulnerability psychopathology. this context, placenta, which is main mother-fetus interface, appears facilitate such convergence, re-directing "stress" signals fetus. Characterizing converging pathways activated by adverse environmental conditions fundamental assess emergence of signatures major disorders, might enable preventive measures populations, and open up new diagnostics, potentially therapeutic approaches disease prevention promotion already pregnancy.

Childhood maltreatment (CM) has well-established consequences for the mental and physical health of exposed individual. Accumulating evidence now suggests that detrimental sequelae CM may be transmitted from one generation to next, thereby extending long-term ramifications early adverse experiences constituting intergenerational continuity in poor outcomes. In this review, current state knowledge on effects maternal exposure is summarized transmission pathways are discussed, specifically direct as well indirect involving variation gestational biology. The review begins with a definition an overview clinical neurobiological offspring generation. intrauterine period biology identified potential time window mechanism transmission, respectively. Background: Exposome research aims to describe and understand the extent which all exposures in human environments may affect our health over lifetime. However, way humans interact with their environment is socially patterned. Failing account for social factors exploring exposome underestimate magnitude of effect or mask inequalities distribution both outcomes. Objectives: We aimed appear literature, manner they are used empirical analyses statistical modeling, considered overall scientific approach. Methods: conducted a scoping review literature using three databases (PubMed, Embase, Web Science) up January 2022. grouped studies based on variables were quantified type frequency mentioned articles. also qualitatively described approach by authors integrate variables. Results: screened 1,001 records, 73 included analysis. Fifty-five (∼75%) as confounders both, wide array represented Individual-level more often found, especially education race/ethnicity, well neighborhood-level deprivation indices. Half hypothesis-free other half, hypothesis-driven latter group, 35 studies, only 8 reported discussed at least one possible mechanism underlying relationship observed between variable outcome. Discussion: Social should be systematic way, considering role structuring specific external internal exposome. Doing so could help mechanisms construction and, potentially, alleviate mitigate emergence new ones. https://doi.org/10.1289/EHP11015

Both Alzheimer's disease and vascular dementia are the result of processes that typically develop over several decades. Population studies have estimated more than half risk for is preventable or at least modifiable through behavioral adaptations. The association between these lifestyle factors most evident exposure in midlife. However, habits formed middle age often reflect a lifetime behavior patterns living conditions. Therefore, individuals who, example, able to maintain healthy diets regular exercise during their years likely benefit from cognition-protective they practiced throughout lives. For numerous adult diseases, significant risks can be traced back early childhood. Suboptimal conditions perinatal period, childhood adolescence increase including stroke, heart disease, insulin resistance, hypertension dementia.
