The formation of new blood vessels, known as angiogenesis, significantly impacts the development multiple types cancer. Consequently, researchers have focused on targeting this process to prevent and treat numerous disorders. However, most existing anti-angiogenic treatments rely synthetic compounds humanized monoclonal antibodies, often expensive or toxic, restricting patient access these therapies. Hence, pursuit discovering new, affordable, less efficient is imperative. Numerous studies propose that natural plant-derived products exhibit sought-after characteristics. objective review delve into properties exhibited by naturally derived flavonoids from plants, along with their underlying molecular mechanisms action. Additionally, we summarize structure, classification, relationship between signaling pathways in plants agents, including main HIF-1α/VEGF/VEGFR2/PI3K/AKT, Wnt/β-catenin, JNK1/STAT3, MAPK/AP-1 pathways. Nonetheless, further research innovative approaches are required enhance bioavailability for clinical application.

Fisetin (Fis), quercetin (Que), and myricetin (Myr) are flavonols with similar structure but different number of hydroxyl groups. The present research focused on the anti-inflammatory effect these three in lipopolysaccharide-stimulated RAW264.7 cells. site group obviously affected their anti-inflammation activity. These suppressed overproduction nitric oxide. Fis showed best activity an inhibition rate 52% at 20 μM. Moreover, reduced levels ROS, TNF-α, IL-6. mechanistic study that they inhibited activation NF-κB MAPK pathways by suppressing phosphorylation IκBα, p65, JNK, ERK, p38, MEK, reducing nuclear translocation p65. In addition, metabolism was examined. results indicated both methylated glucuronidated. Que Myr were mainly transformed into products. This highlights flavonols, particularly Fis, which has potential for prevention or treatment inflammation as adjuvant medicine food additive.

Ficus erecta, a traditional Chinese She Ethnomedicine, has been historically utilized to treat various inflammatory conditions such as arthritis, nephritis, and osteoporosis. However, the underlying mechanisms accounting for its anti-inflammatory activity, well active components, largely remain elusive. The purpose of this research was investigate chemical constituents F. erecta that contribute effects. Coumarins flavones were obtained from 95% EtOH extract using virous column chromatography reversed-phase semipreparative HPLC. structures new compounds elucidated by extensive analysis spectroscopic methods, including HRESIMS, 1D 2D NMR spectra, CD experiments. Cultured macrophage RAW264.7 cells MTT cell viability assay, Griess reagent method, ELISA, Western blot experiments employed evaluate activity related mechanism. Four (1–4) eleven previously identified (5–16) coumarins, together with one (17) six known (18–23) isolated whole plant erecta. Compounds 7 17 significantly reduced nitric oxide (NO) prostaglandin E2 (PGE2) production without cytotoxic Furthermore, proinflammatory cytokines tumor necrosis factor (TNF)-α, interleukin (IL)-1β, IL-6 in concentration-dependent manner. indicated suppressed expression iNOS, COX-2, p-IκBα LPS-stimulated cells. current phytochemical investigations revealed coumarins represent primary exhibit potent properties, linked inhibition NF-κB activation preventing degradation IκBα phosphorylation. These may serve promising candidates treating or certain diseases.

Chrysoeriol is an active ingredient derived from the Chinese medicinal herb (CMH) “Lonicerae japonicae flos” in dried flower bud or bloomed of Lonicera japonica Thunberg. Dermatoses are most common diseases humans, including eczema, acne, psoriasis, moles, and fungal infections, which temporary permanent may be painless painful. Topical corticosteroids widely used Western medicine, but there some side effects when it continuously regularly utilized a large dosage. natural ingredient, nontoxic, without any adverse reactions treatment dermatological conditions. Methods: Nine electronic databases were searched, WanFang Data, PubMed, Science Direct, Scopus, Web Science, Springer Link, SciFinder, China National Knowledge Infrastructure (CNKI), regard to language constraints. The pharmacological activities chrysoeriol Lonicerae flos fight against skin explained evaluated through literature review either vitro vivo studies. Results: decreased mRNA levels proinflammatory cytokines IL-6, IL-1β, TNF-α. These transcriptionally regulated by NF-κB STAT3 combat inflammation. It also showed promising actions treating many ailments wound healing, depigmentation, photoprotection, antiaging. Conclusion: cutaneous route best delivery approach across barrier. However, toxicity, dosage, safety assessments formulation nanochrysoeriol on human epidermis for application must further investigated.

Abstract Glucolipid metabolism disorder (GLMD) is a complex chronic disease characterized by glucose and lipid disorders with diverse etiology rapidly increasing incidence. Many studies have identified the role of flavonoids in ameliorating GLMD, mechanisms related to peroxisome proliferator‐activated receptors, nuclear factor kappa‐B, AMP‐activated protein kinase, (erythroid‐derived 2)‐like 2, transporter type 4, phosphatidylinositol‐3‐kinase/protein kinase B pathway. However, comprehensive summary flavonoid effects on GLMD lacking. This study reviewed roles natural different structures treatment reported globally past 5 years provides reference for developing as drugs treating GLMD.

Inflammation is thought to be a key cause of many chronic diseases and cancer. However, current therapeutic agents control inflammation have limited long-term use potential due various side-effects. This study aimed examine the preventive effects norbergenin, constituent traditional anti-inflammatory recipes, on LPS-induced proinflammatory signaling in macrophages elucidate underlying mechanisms by integrative metabolomics shotgun label-free quantitative proteomics platforms. Using high-resolution mass spectrometry, we identified quantified nearly 3000 proteins across all samples each dataset. To interpret these datasets, exploited differentially expressed conducted statistical analyses. Accordingly, found that production NO, IL1β, TNFα, IL6 iNOS was alleviated norbergenin via suppressed activation TLR2 mediated NFκB, MAPKs STAT3 pathways. In addition, capable overcoming LPS-triggered metabolic reprogramming restrained facilitated glycolysis, promoted OXPHOS, restored aberrant metabolites within TCA cycle. linked its modulation enzymes support activity. Thus, our results uncover regulates inflammatory cascades LPS stimulated exert potential.
