As cancer being the most difficult disease to treat, different kinds of medications and therapeutic approaches have been prominently developed by scientists. For certain families drugs, such as immuno-therapeutics or antibody-drug conjugates, efficient delivery systems are required during administration protect drugs from chemical degradation biological inactivation. Delivery with ability carry therapeutics diagnostic agents both, hold promising potential tackle abnormalities behind cancer. In this context, review provides updated insights on how cyclodextrin-based polymeric nanosystems become an effective treatment approach against Cyclodextrins (CDs) natural oligosaccharides that famously exploited in pharmaceutical research due their exceptional quality entrapping water-insoluble molecules inside hydrophobic core providing enhanced solubility help hydrophilic exterior. Combining properties CDs nanoparticles (PNPs) brings out excellent versatile tunable profiles, thanks submicron-sized PNPs. By introducing significance CD a system, collective discussion binding release mechanisms CD-drug complexation, followed characterization studies has done review. Further, light recent studies, article majorly focuses conveying promoting nanoscale elevates multifunctional advantages can be successfully applied combination therapy theranostics. Moreover, CD-based including CALAA-01, CRLX101, CRLX301, demonstrated improved tumor targeting, reduced side effects, prolonged drug preclinical clinical trials.

Abstract Senescence is a heterogenous and dynamic process in which various cell types undergo cell‐cycle arrest due to cellular stressors. While senescence has been implicated aging many human pathologies, therapeutic interventions remain inadequate the absence of comprehensive set biomarkers context‐dependent manner. Polyphenols have investigated as senotherapeutics both preclinical clinical settings. However, their use hindered by limited stability, toxicity, modest bioavailability, often concentration at target sites. To address these limitations, nanocarriers such polymer nanoparticles lipid vesicles can be utilized enhance efficacy senolytic polyphenols. Focusing on widely studied agents—specifically fisetin, quercetin, resveratrol—we provide concise summaries physical chemical properties, along with an overview findings. We also highlight common signaling pathways potential toxicities associated agents. Addressing challenges linked nanocarriers, we present examples senotherapeutic delivery types, without nanocarriers. Finally, continued research development agents are encouraged reduce undesirable effects different organs. This review underscores need for establishing reliable sets that could assist evaluating effectiveness current future candidates

Abstract Despite progress in cancer treatments, it remains a leading cause of death worldwide. Current chemotherapy is often hampered by side effects, tissue damage, and drug resistance. To overcome these limitations, research exploring new approaches to improve efficacy reduce its effects. A promising strategy involves combining chemotherapeutic drugs with phytochemicals, which are active compounds derived from plants. Nanotechnology plays crucial role delivering often‐hydrophobic drugs, enhancing their bioavailability solubility. This area represents significant convergence nanotechnology both conventional pharmaceuticals botanical compounds. Global studies increasingly demonstrating the potential combination therapies inhibit cell growth laboratory preclinical models. These suggest synergistic or additive effects that lead improved treatment outcomes. article focuses on recent advancements nano‐drug delivery systems, specifically those designed for simultaneous phytochemicals. It explores selection phytochemicals based anticancer properties, outlines design considerations nanocarriers, discusses how physicochemical characteristics influence release kinetics. Additionally, review examines nanoformulations integrate synthetic natural components, highlighting herbal oils medicines treatment.

Piperlongumine, or piplartine (PL), is a bioactive alkaloid isolated from Piper longum L. and potent phytoconstituent in Indian Ayurveda traditional Chinese medicine with lot of therapeutic benefits. Apart all its biological activities, it demonstrates multimodal anticancer activity by targeting various cancer-associated pathways being less toxic to normal cells. According their structure-activity relationship (SAR), the trimethylphenyl ring (cinnamoyl core) 5,6-dihydropyridin-2-(1H)-one (piperdine are responsible for activity. However, has poor intrinsic properties (low aqueous solubility, bioavailability, etc.). As result, pharmaceutical researchers have been trying optimise modify structure PL improve drug-likeness profiles. The present review selected 26 eligible research articles on derivatives published between 2012 2023, followed preferred reporting items systematic reviews meta-analyses (PRISMA) format. We thoroughly summarised potency, mode action, SAR drug chemistry proposed PL-derivatives against different cancer Overall, analyses respect potency drug-ability revealed that substitution methoxy hydroxyl, attachment ligustrazine 4-hydroxycoumarin heterocyclic rings place phenyl rings, like indole at C7-C8 olefin position native can help activity, cell permeability, making them potential leads. Hopefully, large-scale collection critical drug-chemistry will be helpful academic developing potential, less-toxic cost-effective used cancers.
