Numerous studies have shown that berberine and its derivatives demonstrate important anti-tumor effects. However, the specific underlying mechanism remains unclear. Therefore, based on systems pharmacology, this review summarizes information available effects of derivatives. This pharmacology systematically examines action potential derivatives, used in treatment complex cancers, at molecular, cellular, organismic levels. Moreover, with further in-depth investigations toxicity efficacy, could be drugs cancer therapy, improved clinical efficacy safety.

Osteomyelitis, as a severe bone disease caused by bacterial infection, can result in lifelong disability or fatal sepsis. Considering that the infection is stubborn and deep-sited tissue, situ rapid treatments for osteomyelitis remain significant challenge. Herein, we prepare an ultrasound (US)-activated single-atom catalyst consists of Au nanorod (NRs)-actuated single-atom-doped porphyrin metal–organic framework (HNTM-Pt@Au) red cell membrane (RBC), which efficiently treat methicillin-resistant Staphylococcus aureus (MRSA)-infected under US. Besides outstanding performance field photocatalysis, find single atoms (such Pt, Au, Cu) also improve sonocatalytic ability sonosensitizer. Due to strong electron-trapping oxygen adsorption capacity, Pt atom endows RBC-HNTM-Pt@Au with excellent activity. It shows antibacterial efficiency 99.9% toward MRSA 15 min US irradiation. Meanwhile, be propelled directionally thus dynamically neutralize secreted toxins. The MRSA-infected rat tibia was successfully treated, negligible loss, reduced inflammation response, great biocompatibility. This work presents efficient sonodynamic therapy treatment deep tissue infections via multifunctional catalyst.

S. aureus is resistant to various first-line antibiotics, and seeking multifarious strategies aimed at effective control of antibiotic-resistant behavior urgently needed. Here, we report a two-component directed self-assembly mode: the phytochemicals berberine cinnamic acid can directly self-assemble into nanoparticles (NPs) displaying good bacteriostastic activity. Compared with several obtained nanostructures have better inhibitory effect on multidrug-resistant (MRSA) stronger ability for biofilm removal. These qualities are attributed fact that organic assemblies first spontaneously adhere surface bacteria, infiltrate cell, then lead converging attack against MRSA; thereafter, multipath bactericidal mechanisms NPs MRSA found by both transcriptomic analysis quantitative Polymerase Chain Reaction analysis. Moreover, when combined spectral data single crystal X-ray diffraction, NPs' mechanism governed hydrogen bonds π–π stacking interactions clearly elucidated. non-covalent induce formation butterfly-like one-dimensional self-assembled units finally layered three-dimensional spatial configuration. In addition, biocompatibility tests show nonhemolytic little toxicity in vitro vivo. This mode offer new perspective toward design biocompatible antimicrobial nanomedicines clinical translation.

Effective drug delivery and prevention of postoperative recurrence are significant challenges for current glioblastoma (GBM) treatment. Poor is mainly due to the presence blood–brain barrier (BBB), primarily resistance GBM cells chemotherapeutic drugs an immunosuppressive microenvironment. Herein, a biomimetic nanodrug platform based on endogenous exosomes that could efficiently target brain without targeting modifications co-deliver pure nanomicelles immune adjuvants safe efficient chemo-immunotherapy against prepared. Inspired by self-assembly technology small molecules, tanshinone IIA (TanIIA) glycyrrhizic acid (GL), which inhibitors signal transducers activators transcription 3 from traditional Chinese medicine (TCM), self-assembled form TanIIA-GL (TGM). Endogenous serum selected coat nanomicelles, CpG oligonucleotides, agonists Toll-like receptor 9, anchored exosome membrane obtain loaded with TCM (CpG-EXO/TGM). Methicillin-resistant Staphylococcus aureus (MRSA)-infected skin wounds have caused a variety of diseases and seriously endanger global public health. Therefore, multidimensional strategies are urgently to find antibacterial dressings combat bacterial infections. Antibacterial hydrogels considered potential wound dressing, while their clinical translation is limited due the unpredictable risks high costs carrier excipients. it found that natural star anti-inflammatory phytochemicals baicalin (BA) sanguinarine (SAN) can directly self-assemble through non-covalent bonds such as electrostatic attraction, π-π stacking, hydrogen bonding form carrier-free binary small molecule hydrogel. In addition, BA-SAN gel exhibited synergistic inhibitory effect on MRSA. And its plasticity injectability allowed be applied dressing. Due matched physicochemical properties therapeutic effects, inhibit virulence factors, alleviate inflammation, promote healing, has good biocompatibility. The current study not only provided an hydrogel with value but also opened up new prospects designed originated from clinically used small-molecule phytochemicals.

Bacterial infections, such as bacterial keratitis (BK) and subcutaneous abscess, pose significant challenges to global healthcare. High-entropy MXenes (HE MXenes) used confer desirable properties with exposed active sites high-entropy atomic layers, whose potential application the field biomedicine remains be explored. Herein, monolayer HE fabricated by implementing transition metals entropy low Gibbs free energy fill gap biocatalytic performance non-high-entropy MXenes. endowed extremely strong oxidase mimic activity (Km = 0.227 mm) photothermal conversion efficiency (65.8%) second near-infrared (NIR-II) biowindow increases. However, the bioavailability extract raw herbs bioactive compounds is poor because their water solubility. In order to overcome solubility issues, electrospinning technology can offer delivery alternative resolve them.
