Abstract Dielectrophoresis (DEP) particle separation has label‐free, well‐controllable, and low‐damage merits. Sidewall microelectrodes made of liquid metal alloy (LMA) inherits the additional advantage thick electrodes to generate impactful DEP force. However, existing LMA electrode‐based devices lack ability integrate large‐array in a compact footprint, severely limiting flow rate thus throughput. Herein, facile versatile method is proposed high‐density microfluidic devices, taking passive control capillary burst valves (CBVs). CBVs with carefully designed pressures are co‐designed channels, allowing self‐assembly electrode array through simple hand‐push injection. The arrayed configuration brings accumulative deflection effect. Specifically, fabricated 5000 pairs sidewall chip demonstrted achieve ten times higher throughput deflection. 5000‐electrode‐pair device applied successfully separate four mixed samples, including human peripheral blood mononuclear cells A549 70 µL min −1 . It envisioned that this work can greatly facilitate fabrication offer robust platform for applications.

Abstract Single‐cell mass spectrometry (MS) is significant in biochemical analysis and holds great potential biomedical applications. Efficient sample preparation like sorting (i.e., separating target cells from the mixed population) desalting moving off non‐volatile salt solution) urgently required single‐cell MS. However, traditional methods suffer complicated operation with various apparatus, or insufficient performance. Herein, a one‐step strategy by leveraging label‐free impedance flow cytometry (IFC) based microfluidics proposed. Specifically, IFC framework to characterize sort single‐cells adopted. Simultaneously sorting, cell transferred local high‐salinity buffer MS‐compatible solution. In this way, are achieved collected can be directly fed for MS analysis. A high efficiency (>99%), cancer purity (≈87%), whole workflow of impedance‐based separation normal (MCF‐10A) (MDA‐MB‐468) verified. As standalone module, microfluidic chip compatible variety methods, envisioned provide new paradigm efficient preparation, further multi‐modal electrical metabolic) characterization single‐cells.

Abstract Reflecting various physiological states and phenotypes of single cells, intrinsic biophysical characteristics (e.g., mechanical electrical properties) are reliable important, label‐free biomarkers for characterizing cells. Impedance flow cytometry (IFC) has been demonstrated to be an efficient tool for label-free bacterial investigation obtain the electrical properties in real time. However, accurate differentiation of different species bacteria by IFC technology remains a challenge owing insignificant differences data. Here, we developed convolutional neural networks (ConvNet) deep learning approach enhance accuracy and efficiency toward distinguishing various bacteria. First, more than 1 million sets impedance data (comprising 42 characteristic features each set) groups were trained ConvNet model. To improve analysis, Spearman correlation coefficient mean decrease random forest algorithm introduced eliminate feature interaction extract opacity related wall membrane structure as predominant differentiation. Moreover, 25 optimized selected with accuracies >96% three (bacilli, cocci, vibrio) >95% two bacilli (Escherichia coli Salmonella enteritidis), compared machine algorithms (complex tree, linear discriminant, K-nearest neighbor algorithms) maximum 76.4%. Plant pathogen zoospores play a vital role in the transmission of several significant plant diseases, with their early detection being important for effective management. Current methods involve labour-intensive specimen collection and laboratory testing, lacking real-time feedback capabilities. Methods that can be deployed field remotely addressed are required. In this study, we have developed an innovative zoospore-sensing device by combining microfluidic sampling system cytometer incorporating chemotactic response as means to selectively detect motile spores. Spores Phytophthora cactorum were guided swim up channel following gradient attractant. They then detected transient change impedance when they passed between pair electrodes. Single-zoospore was demonstrated signal-to-noise ratios ~17 carrying flow used ~5.9 induced into attractants. This work provides solution selective, sensitive zoospores.
