Curcumin, a yellow pigment in the Indian spice Turmeric (Curcuma longa), which is chemically known as diferuloylmethane, was first isolated exactly two centuries ago 1815 by German Scientists, Vogel and Pelletier. However, according to pubmed database, study on its biological activity an antibacterial agent published 1949 Nature clinical trial reported The Lancet 1937. Although current database indicates almost 9000 publications curcumin, until 1990 there were less than 100 papers this nutraceutical. At molecular level, multitargeted has been shown exhibit anti-inflammatory through suppression of numerous cell signalling pathways including NF-κB, STAT3, Nrf2, ROS COX-2. Numerous studies have indicated that curcumin highly potent antimicrobial be active against various chronic diseases types cancers, diabetes, obesity, cardiovascular, pulmonary, neurological autoimmune diseases. Furthermore, compound also synergistic with other nutraceuticals such resveratrol, piperine, catechins, quercetin genistein. To date, over different trials completed clearly show safety, tolerability effectiveness humans. more populations are necessary prove potential This review's primary focus lessons learnt about from trials.

Turmeric is a curry spice that originated from India, which has attracted great interest in recent decades because it contains bioactive curcuminoids (curcumin, demethoxycurcumin, and bisdemethoxycurcumin). Curcumin (1,7-bis-(4-hydroxy-3-methoxyphenyl)-hepta-1,6-diene-3,5-dione), lipophilic polyphenol may work as an anticancer, antibiotic, anti-inflammatory, anti-aging agent suggested by several vitro, vivo studies clinical trials. However, poor aqueous solubility, bioavailability, pharmacokinetic profiles limit curcumin’s therapeutic usage. To address these issues, curcumin formulations have been developed. suboptimal sample preparation analysis methodologies often hamper the accurate evaluation of bioactivities their efficacy. This review summarizes research on biological, pharmaceutical, analytical aspects curcumin. Various formulation techniques corresponding trials outcomes are discussed. A detailed comparison different (ultrasonic, pressurized liquid extraction, microwave, reflux) (FT-IR, FT-NIR, FT-Raman, UV, NMR, HPTLC, HPLC, LC-MS/MS) used for extraction quantification matrices, presented. Application optimal preparation, chromatographic separation, detection will significantly improve assessment biological activities curcuminoids.

This review addresses the oncopharmacological properties of curcumin at molecular level. First, interactions between and its targets are addressed on basis curcumin's distinct chemical properties, which include H-bond donating accepting capacity β-dicarbonyl moiety phenylic hydroxyl groups, methoxy ethers, multivalent metal nonmetal cation binding high partition coefficient, rotamerization around multiple C–C bonds, ability to act as a Michael acceptor. Next, in vitro stability is elaborated context susceptibility photochemical modification degradation (e.g., alkaline hydrolysis). Specific degradatory pathways provided, mainly entail radical-based intermediates, catabolites identified. The implications (photo)chemical instability light pharmaceutical preparations, use analogues, implementation nanoparticulate drug delivery systems. Furthermore, pharmacokinetics most important products detailed poor bioavailability. Particular emphasis placed xenobiotic phase I II metabolism well excretion intestines (first pass), liver (second other organs addition metabolites their systemic clearance. Lastly, summary provided clinical pharmacodynamics followed by account direct targets, whereby phenotypical/biological changes induced cancer cells upon completion curcumin-triggered signaling cascade(s) framework hallmarks cancer. ErbB family receptors, protein kinase C, enzymes involved prostaglandin synthesis, vitamin D receptor, DNA.

Curcumin is a natural compound of Curcuma longa L. and has shown many pharmacological activities such as anti-inflammatory, anti-oxidant in both preclinical clinical studies. Moreover, curcumin hepatoprotective, neuroprotective protects against myocardial infarction. Particularly, also demonstrated favorite anticancer efficacy. But limiting factors its extremely low oral bioavailability hampers application therapeutic agent. Therefore, technologies have been developed applied to overcome this limitation. This review described the main physicochemical properties summarized recent studies design development delivery systems for enhance solubility bioavailability, including liposomes, nanoparticles polymeric micelles, phospholipid complexes, microemulsions.

The yellow pigment curcumin, extracted from turmeric, is a renowned polyphenol with broad spectrum of health properties such as antioxidant, anti-inflammatory, anti-cancer, antidiabetic, hepatoprotective, anti-allergic, anti-dermatophyte, and neuroprotective. However, these are followed by poor pharmacokinetic profile which compromises its therapeutic potential. association low absorption the small intestine extensive reductive conjugative metabolism in liver dramatically weakens oral bioavailability. Several strategies inhibition curcumin adjuvants well novel solid liquid delivery systems have been tried to counteract rapid elimination body. Some drug deliveries can successfully enhance solubility, extending residence plasma, improving cellular uptake.

Abstract Curcumin, an active polyphenol of the golden spice turmeric, is a highly pleiotropic molecule with potential to modulate biological activity number signaling molecules. Traditionally, this has been used in Asian countries treat such human ailments as acne, psoriasis, dermatitis, and rash. Recent studies have indicated that curcumin can target newly identified pathways including those associated microRNA, cancer stem cells, autophagy. Extensive research from preclinical clinical delineated molecular basis for pharmaceutical uses against cancer, pulmonary diseases, neurological liver metabolic autoimmune cardiovascular numerous other chronic diseases. Multiple safety efficacy animals rodents, monkeys, horses, rabbits, cats provided solid evaluating its humans. To date, more than 65 trials curcumin, which included 1000 patients, completed, many 35 are underway. Curcumin now supplement several United States, India, Japan, Korea, Thailand, China, Turkey, South Africa, Nepal, Pakistan. In review, we provide evidence various © 2013 BioFactors, 39(1):2–13,
