Background Mental disorders have a high comorbidity with cardiovascular disease (CVD), but the causality between them has not been fully appreciated. Objective This study aimed to systematically explore bidirectional two broad categories of diseases. Methods We conducted Mendelian randomisation (MR) and multivariable MR (MVMR) analyses evaluate potential causal links 10 mental disorders, use antidepressants 7 CVDs. Findings discovered that major depressive disorder (MDD), attention-deficit/hyperactivity (ADHD) insomnia exhibit connections elevated risks or more Moreover, is linked heightened each CVD. Each distinct CVD correlated greater probability taking antidepressants. Our MVMR analysis demonstrated elevation respective across all conditions. includes arrhythmias (OR: 1.28), atrial fibrillation 1.44), coronary artery 1.16), hypertension heart failure stroke 1.44) entire group 1.35). However, MDD itself was risk any Conclusions The findings our indicate MDD, ADHD may increase highlight utilisation as an independent factor for CVD, thus explaining influence on through mediating effects Clinical implications When treating patients antidepressants, it necessary take into consideration beneficial detrimental

The high comorbidity of major depressive disorder (MDD) with other diseases has been well-documented. However, the pairwise causal connections for MDD comorbid networks are poorly characterized. We performed Phenome-wide Mendelian randomization (MR) analyses to explore bidirectional associations between (N = 807,553) and 877 common from FinnGen datasets 377,277). inverse variance weighting method was primary technique, methods (weighted median MR-Egger) were used sensitivity analyses. Our MR showed that genetic liability is causally associated risks 324 disease phenotypes (average b: 0.339), including 46 psychiatric behavioral disorders 0.618), 18 neurological 0.348), 44 respiratory 0.345), 40 digestive 0.281), circulatory 0.237), 37 genitourinary 0.271), 66 musculoskeletal connective 0.326), 22 endocrine 0.302), others. In a reverse analysis, total 51 components predisposing various risk 0.086), 5 infectious 0.056), 11 0.106), 14 oncological 0.108), 0.114). Bidirectional identified 15 diseases. For most analyses, little evidence heterogeneity pleiotropy detected. findings confirmed extensive significant role predisposition in contributing human phenotypes, which more pronounced than those seen analysis influences on MDD.

Abstract Background Existing evidence suggests that alterations in the gut microbiome are closely associated with major depressive disorder (MDD). We aimed to reveal causal relationships between MDD and various microbial taxa gut. Methods used two-sample Mendelian randomization (TSMR) explore bidirectional effects microbiota MDD. The genome-wide association studies summary results of were obtained from two large consortia, MibioGen consortium Dutch Microbiome Project, which we analyzed separately. Results Our TSMR analysis identified 10 bacterial protective against MDD, including phylum Actinobacteria , order Clostridiales family Bifidobacteriaceae (OR: 0.96 ∼ 0.98). Ten an increased risk phyla Firmicutes Proteobacteria class genus Alistipes 1.01 1.09). On other hand, may decrease abundance 12 taxa, families Defluviitaleaceae 0.63 0.88). increase 8 Bacteroidetes genera Parabacteroides Bacteroides 1.12 1.43). Type 2 diabetes (T2D) is commonly co-morbid with Alzheimer's disease (AD). However, it remains unclear whether T2D itself or the antidiabetic drug metformin contributes to progression of AD. This study aimed investigate overall and independent effects use on risk Summary genome-wide association datasets were utilized for Mendelian randomization (MR) multivariable MR (MVMR) analyses, including ones (N = 455,017), 456,276), AD 453,733). Additionally, using proportional imbalance method, we analyzed AD-related adverse events in FDA Adverse Event Reporting System (FAERS) database (covering Q1 2004 Q2 2024). Our two-sample analysis indicated that not associated (OR: 1.03, CI: 0.99-1.08, P 0.128). while statistically significant, genetic signature exposure demonstrated a trend toward an increased 1.05, 1.00-1.09, 0.053). Interestingly, MVMR analysis, which evaluates T2D, found robust decrease 0.82, 0.68-0.98, 0.031), was higher 1.26, 1.06-1.50, 9.45E-3). In FAERS database, total 228,283 metformin-related event reports from 67,742 cases found. For as target event, signal reported 29 (ROR: 0.83, 95 % 0.58-1.19, 0.3126). reveals opposite causal These findings highlight importance assessing when prescribing patients T2D.
