Abstract Research into the biophysical properties of deoxyribonucleic acid (DNA) and mechanisms underlying genetic mutations has undergone marked advancements. The intriguing nature resulting from retroviral DNA insertion garnered considerable attention. Whether these are random or region-specific, distribution patterns mutation sites have been focus numerous research endeavours. This mechanism originates interactions between host pre-integration complex (PIC), comprising an integrase enzyme that facilitates its incorporation DNA. Human papillomavirus associated anogenital cancers are a significant global burden. The detection of biomarkers (circulating tumour DNA; ctDNA or circulating HPV cHPV DNA) in blood referred to as "liquid biopsy" may support the early diagnosis and monitoring affected individuals. A systematic review, including meta-analysis studies available literature on utilization DNA diagnostic, predictive, biomarker tests was performed following criteria PRISMA. total 31 were eligible for review; 20 used cervical cancers; 7 cancer; 5 anal no vulva, vaginal penile cancer available. identified low sensitivity (0.36) high specificity (0.96) diagnostic cancer. Abstract Cell-free DNA (cfDNA) sequencing has demonstrated great potential for early cancer detection. However, most large-scale studies have focused only on either targeted methylation sites or whole-genome sequencing, limiting comprehensive analysis that integrates both epigenetic and genetic signatures. In this study, we present a platform enables simultaneous of methylation, copy number, fragmentomic patterns cfDNA in single assay. Using total 950 plasma (361 healthy 589 cancer) 240 tissue samples, demonstrate multifeature signature ensemble (CSE) classifier integrating all features outperforms single-feature classifiers. At 95.2% specificity, the detection sensitivity with models was 77.2%, 61.4%, 60.5%, respectively, but significantly increased to 88.9% CSE ( p value < 0.0001). Cell-free (cf) HPV DNA detection in blood samples may serve as a monitoring tool for cancer.
