With the aging global population, type 2 diabetes mellitus (T2DM) and osteoporosis(OP) are becoming increasingly prevalent. Diabetic osteoporosis (DOP) is a metabolic bone disorder characterized by abnormal tissue structure reduced strength in patients with diabetes. Studies have revealed close association among diabetes, increased fracture risk, disturbances iron metabolism. This review explores concept of ferroptosis, non-apoptotic cell death process dependent on intracellular iron, focusing its role DOP. Iron-dependent lipid peroxidation, particularly impacting pancreatic β-cells, osteoblasts (OBs) osteoclasts (OCs), contributes to The intricate interplay between dysregulation, which comprises deficiency overload, DOP has been discussed, emphasizing how excessive accumulation triggers ferroptosis concise overview highlights need understand complex relationship T2DM OP, ferroptosis. aimed elucidate pathogenesis provide prospective for future research targeting interventions field

The ovarian microenvironment is critical for follicular development and oocyte maturation. Maternal conditions, including polycystic ovary syndrome (PCOS), endometriosis, aging, may compromise the microenvironment, development, quality. Chronic low-grade inflammation can induce oxidative stress tissue fibrosis in ovary. In PCOS, pro-inflammatory cytokine levels are often elevated fluids. women with obesity hyperandrogenemia insulin resistance chronic inflammation, thereby disrupting by increasing stress. endometrioma-derived iron overload stress, leading to ferroptosis fibrosis. inflammatory aging (inflammaging), senescent cells secrete senescence-associated secretory phenotype factors, causing Therefore, controlling would present a novel therapeutic strategy improving minimizing dysfunction.

Abstract STUDY QUESTION Does ovarian ferroptosis play an active role in the development of polycystic ovary syndrome (PCOS)? SUMMARY ANSWER Increased was present PCOS ovaries and inhibition with ferrostatin-1 (Fer-1) ameliorated morphology anovulation. WHAT IS KNOWN ALREADY Programmed cell death plays a fundamental follicle development. However, types mechanisms involved are yet to be elucidated. Ferroptosis is recently discovered iron-dependent programmed death. Impaired iron metabolism have been observed women PCOS, main cause anovulatory infertility. Additionally, previous studies reported that abnormal expression noncoding RNA may promote immortalized granulosa lines. little known about whether increased there insufficient direct evidence for underlying mechanism. Moreover, effect Fer-1 remains unclear. DESIGN, SIZE, DURATION evaluated human cells (hGCs) from non-PCOS (n = 6–16) 7–18) patients. Intervertebral disc degeneration (IVDD) is a common chronic orthopaedic disease in orthopaedics that imposes heavy economic burden on people and society. Although it well established IVDD associated with genetic susceptibility, ageing obesity, its pathogenesis remains incompletely understood. Previously, was thought to occur because of excessive mechanical loading leading destruction nucleus pulposus cells (NPCs), but studies have shown much more complex process inflammation, metabolic factors NPCs death can involve all parts the disc, characterized by causing extracellular matrix (ECM) degradation. The damage pattern like some programmed cell death, suggesting death. apoptosis pyroptosis been studied IVDD, intervertebral still not be fully elucidated using only traditional modalities. With increasing research, new modes PANoptosis, ferroptosis senescence found closely related degeneration. Among these, PANoptosis combines essential elements pyroptosis, necroptosis form highly coordinated dynamically balanced inflammatory process. Furthermore, we believe may also crosstalk senescence. Therefore, review progress research multiple deaths provide guidance for clinical treatment.

ABSTRACT Ovarian endometrioma (OEM), a particularly severe form of endometriosis, is an oestrogen‐dependent condition often associated with pain and infertility. The mechanisms by which OEM impairs fertility, through its direct impact on oocyte‐cumulus cell (CC) communication related pathways, remain poorly understood. This study investigates the oocyte‐CC explores melatonin's therapeutic potential. We used mouse model employed ovarian transcriptome gene set enrichment analyses to identify disrupted alongside phalloidin staining for cytoskeletal analysis, gap junction coupling analysis intercellular communication, mitochondrial function assessments cellular metabolism. Our results showed that significantly steroidogenesis cumulus function, leading increased apoptosis, transzonal projections (TZPs), impaired antioxidant transfer oocytes. culminates in oxidative stress, dysfunction, compromised ATP production. oocytes also exhibited abnormalities, including DNA damage, maturation defects, spindle assembly disruptions, aneuploidy. identifies TZPs as key pathological feature highlights potential restore mitigate improve reproductive outcomes.

Endometriosis, the most prevalent cause of infertility, is associated with anatomical distortion leading to adhesions and fibrosis, as well endocrine abnormalities immune disorders. This review discusses mechanisms underlying endometriosis-related infertility. Firstly, alterations in hypothalamic-pituitary-ovarian axis lead secretion gonadotropins steroid hormones, adverse effects on ovulation implantation, fertility decline. Secondly, dysregulation hypothalamic-pituitary-adrenal induces elevated serum cortisol prolactin levels patients endometriosis, accounting for its regulation stress, depression, anxiety. Abnormal interactions between endometrial cells system change local microenvironment, resulting epithelial-mesenchymal transition inflammation. Activated epithelial cells, stromal immunocytes produce various chemokines, cytokines, or autoantibodies, creating an unfavorable environment embryo implantation. These findings suggest that spectrum play a crucial role Thirdly, oxidative stress has ovarian reserve subsequent embryonic development, predicting another promising strategy An unbalanced redox state, including impaired mitochondrial function, dysregulated lipid metabolism, iron-induced generates pro-oxidative which negatively impacts oocyte quality sperm viability. Thus, updated understanding involved this disease will help develop effective strategies manage

Ferroptosis is a novel form of cell death precisely regulated by iron metabolism, antioxidant processes, and lipid metabolism that plays an irreplaceable role in the development many diseases. Musculoskeletal disorders (MSKs), including osteoporosis, osteoarthritis, rheumatoid arthritis, intervertebral disc degeneration, sarcopenia, rhabdomyolysis, have become one most common causes disability major burden on public health social care systems. The mechanism ferroptosis MSKs has recently been elucidated. In this review, we briefly introduce illustrate pathological roles with focus how can be exploited as promising treatment strategy. Notably, because toxicity compounds inhibit or induce other organs largely unknown, appears to double-edged sword. We point out more research needed future verify therapeutic effects based MSKs.
