Abstract Excess body weight (EBW) increases the risk of colorectal cancer (CRC) and is linked to lower colonoscopy compliance. Here, we extensively analyzed 981 metagenome samples from multiple cohorts pinpoint specific microbial signatures their potential capability distinguishing EBW patients with CRC. The gut microbiome displayed considerable variations between lean We identify 44 37 distinct multi-kingdom species differentiating CRC controls in populations, respectively. Objective To profile gut microbiome-associated metabolites in serum and investigate whether these could distinguish individuals with colorectal cancer (CRC) or adenoma from normal healthy individuals. Design Integrated analysis of untargeted metabolomics by liquid chromatography-mass spectrometry metagenome sequencing paired faecal samples was applied to identify significantly altered abundance patients CRC adenoma. The ability discriminate between tested targeted metabolomic analysis. A model based on established evaluated an independent validation cohort. Results In total, 885 were both adenoma, including eight (GMSM panel) that reproducibly detected accurately discriminated samples. GMSM panel-based predict yielded area under the curve (AUC) 0.98 (95% CI 0.94 1.00) modelling cohort AUC 0.92 (83.5% sensitivity, 84.9% specificity) superior clinical marker carcinoembryonic antigen among within (AUC vs 0.72) also showed promising diagnostic accuracy for adenomas (AUC=0.84) early-stage (AUC=0.93). Conclusion Gut microbiome reprogramming is associated alterations metabolome, GMSMs have potential applications detection.

Abstract Despite recent progress in our understanding of the association between gut microbiome and colorectal cancer (CRC), multi-kingdom dysbiosis CRC across cohorts is unexplored. We investigated four-kingdom microbiota alterations using metagenomic datasets 1,368 samples from 8 distinct geographical cohorts. Integrated analysis identified 20 archaeal, 27 bacterial, fungal 21 viral species for each single-kingdom diagnostic model. However, data revealed superior accuracy models constructed with markers, particular addition species. Specifically, 16 markers including 11 4 1 archaeal feature, achieved good performance diagnosing patients (area under receiver operating characteristic curve (AUROC) = 0.83) maintained 3 independent Coabundance ecological network associations bacterial species, such as Talaromyces islandicus Clostridium saccharobutylicum . Using metagenome shotgun sequencing data, predictive power microbial functional potential was explored elevated D-amino acid metabolism butanoate were observed CRC. Interestingly, model based on EggNOG genes high (AUROC 0.86). Collectively, findings uncovered CRC-associated common demonstrate applicability tools and, potentially, therapeutic targets treatment

Abstract Associations between gut microbiota and colorectal cancer (CRC) have been widely investigated. However, the replicable markers for early-stage adenoma diagnosis across multiple populations remain elusive. Here, we perform an integrated analysis on 1056 public fecal samples, to identify adenoma-associated microbial early detection of CRC. After adjusting potential confounders, Random Forest classifiers are constructed with 11 discriminate from control (area under ROC curve (AUC) = 0.80), 26 CRC (AUC 0.89), respectively. Moreover, validate in two independent cohorts achieving AUCs 0.78 0.84, Functional reveals that altered microbiome is characterized increased ADP- l -glycero-beta- d -manno-heptose biosynthesis elevated menaquinone-10 These findings validated a newly-collected cohort 43 samples using quantitative real-time PCR. This work proves validity adenoma-specific multi-populations, which would contribute treatment

Optical sensors, with great potential to convert invisible bioanalytical response into readable information, have been envisioned as a powerful platform for biological analysis and early diagnosis of diseases. However, the current extraction sensing data is basically processed via series complicated time-consuming calibrations between samples reference, which inevitably introduce extra measurement errors potentially annihilate small intrinsic responses. Here, we proposed experimentally demonstrated calibration-free sensor achieving high-precision biosensing detection, based on an optically controlled terahertz (THz) ultrafast metasurface. Photoexcitation silicon bridge enables resonant frequency shifting from 1.385 0.825 THz reaches maximal phase variation up 50° at 1.11 THz. The typical environmental are completely eliminated in theory by normalizing Fourier-transformed transmission spectra ultrashort time delays 37 ps, resulting extremely robust device monitoring cancerous process gastric cells. We believe that our sensors high precision advantages can extend their implementation study dynamics may inspire considerable innovations field medical devices nondestructive detection.

Colorectal cancer (CRC) is the most common of digestive system with high mortality and morbidity rates. Gut microbiota found in intestines, especially colorectum, has structured crosstalk interactions host that affect several physiological processes. The gut include CRC-promoting bacterial species, such as Fusobacterium nucleatum, Escherichia coli, Bacteroides fragilis, CRC-protecting Clostridium butyricum, Streptococcus thermophilus, Lacticaseibacillus paracasei, which along other microorganisms, viruses fungi, play critical roles development CRC. Different features are identified patients early-onset CRC, combined different patterns between fecal intratumoral microbiota. may be beneficial diagnosis treatment CRC; some bacteria serve biomarkers while others regulators chemotherapy immunotherapy. Furthermore, metabolites produced by essential CRC cells. Harmful primary bile acids short-chain fatty acids, whereas others, including ursodeoxycholic acid butyrate, impede tumor progression. This review focuses on its metabolites, their potential development, diagnosis,

Abstract The perturbations of the gut microbiota and metabolites are closely associated with progression inflammatory bowel disease (IBD). However, inconsistent findings across studies impede a comprehensive understanding their roles in IBD potential as reliable diagnostic biomarkers. To address this challenge, here we comprehensively analyze 9 metagenomic 4 metabolomics cohorts from different populations. Through cross-cohort integrative analysis (CCIA), identify consistent characteristic commensal microbiota. Especially, three bacteria, namely Asaccharobacter celatus , Gemmiger formicilis Erysipelatoclostridium ramosum which rarely reported IBD. Metagenomic functional reveals that essential gene Two-component system pathway, linked to fecal calprotectin, implicated Metabolomics shows 36 identified significant differences, while these still unknown. further elucidate relationship between metabolites, construct multi-omics biological correlation (MOBC) maps, highlights microbial biotransformation deficiencies alterations aminoacyl-tRNA synthetases. Finally, biomarkers for diagnosis, validated multiple global (AUROC values ranging 0.92 0.98). Our results offer valuable insights resource developing mechanistic hypotheses on host-microbiome interactions

Colorectal cancer (CRC) is the third most common malignant tumor worldwide. The incidence and mortality rates of CRC have been increasing in China, possibly due to economic development, lifestyle, dietary changes. Evidence suggests that gut microbiota plays an essential role tumorigenesis CRC. Gut dysbiosis, specific pathogenic microbes, metabolites, virulence factors, microbial carcinogenic mechanisms contribute initiation progression biomarkers potential translational applications screening early diagnosis. microbiota-related interventions could improve anti-tumor therapy's efficacy severe intestinal toxic effects. Chinese researchers made many achievements relationship between CRC, although some challenges remain. This review summarizes current evidence from China on mainly including characteristics, especially Fusobacterium nucleatum Parvimonas micra, which identified be enriched patients; pathogens such as F. enterotoxigenic Bacteroides fragilis, P. scientists extensively studied; diagnostic nucleatum; therapeutic effects, microecological agents represented by certain Lactobacillus strains, fecal transplantation, traditional medicines Berberine Curcumin. More efforts should focused exploring underlying pathogenesis providing novel preventive strategies.
