Intratumoral pathogens can contribute to cancer progression and affect therapeutic response. Fusobacterium nucleatum, a core pathogen of colorectal (CRC), is an important cause low efficacy metastasis. Thus, the modulation intratumoral may provide target for therapy metastasis inhibition. Herein, we propose F. nucleatum-modulating strategy enhancing CRC inhibiting lung by designing antibacterial nanoplatform (Au@BSA-CuPpIX), which produced reactive oxygen species (ROS) under ultrasound exhibited strong activity. Importantly, Au@BSA-CuPpIX reduced levels apoptosis-inhibiting proteins thereby ROS-induced apoptosis. In vivo results demonstrated that effectively eliminated nucleatum enhance sonodynamic (SDT) orthotopic inhibit Notably, entrapped gold nanoparticles phototoxicity metalloporphyrin accumulated in skin during tumor treatment, preventing severe inflammation damage skin. Therefore, this study proposes elimination effect SDT, thus providing promising paradigm improving treatment with fewer toxic side effects promoting clinical translational potential SDT.

Cancer-microbe associations have been explored for centuries, but cancer-associated fungi rarely examined. Here, we comprehensively characterize the cancer mycobiome within 17,401 patient tissue, blood, and plasma samples across 35 types in four independent cohorts. We report fungal DNA cells at low abundances many major human cancers, with differences community compositions that differ among types, even when accounting technical background. Fungal histological staining of tissue microarrays supported intratumoral presence frequent spatial association macrophages. Comparing communities matched bacteriomes immunomes revealed co-occurring bi-domain ecologies, often permissive, rather than competitive, microenvironments distinct immune responses. Clinically focused assessments suggested prognostic diagnostic capacities mycobiomes, stage I synergistic predictive performance bacteriomes.

Abstract The perturbations of the gut microbiota and metabolites are closely associated with progression inflammatory bowel disease (IBD). However, inconsistent findings across studies impede a comprehensive understanding their roles in IBD potential as reliable diagnostic biomarkers. To address this challenge, here we comprehensively analyze 9 metagenomic 4 metabolomics cohorts from different populations. Through cross-cohort integrative analysis (CCIA), identify consistent characteristic commensal microbiota. Especially, three bacteria, namely Asaccharobacter celatus , Gemmiger formicilis Erysipelatoclostridium ramosum which rarely reported IBD. Metagenomic functional reveals that essential gene Two-component system pathway, linked to fecal calprotectin, implicated Metabolomics shows 36 identified significant differences, while these still unknown. further elucidate relationship between metabolites, construct multi-omics biological correlation (MOBC) maps, highlights microbial biotransformation deficiencies alterations aminoacyl-tRNA synthetases. Finally, biomarkers for diagnosis, validated multiple global (AUROC values ranging 0.92 0.98). Colorectal cancer (CRC) is the third most common malignant tumor worldwide. The incidence and mortality rates of CRC have been increasing in China, possibly due to economic development, lifestyle, dietary changes. Evidence suggests that gut microbiota plays an essential role tumorigenesis CRC. Gut dysbiosis, specific pathogenic microbes, metabolites, virulence factors, microbial carcinogenic mechanisms contribute initiation progression biomarkers potential translational applications screening early diagnosis. microbiota-related interventions could improve anti-tumor therapy's efficacy severe intestinal toxic effects. Chinese researchers made many achievements relationship between CRC, although some challenges remain. This review summarizes current evidence from China on mainly including characteristics, especially Fusobacterium nucleatum Parvimonas micra, which identified be enriched patients; pathogens such as F. enterotoxigenic Bacteroides fragilis, P. scientists extensively studied; diagnostic nucleatum; therapeutic effects, microecological agents represented by certain Lactobacillus strains, fecal transplantation, traditional medicines Berberine Curcumin.
