Aging and various age-related diseases are associated with reductions in melatonin secretion, proinflammatory changes the immune system, a deteriorating circadian sirtuin-1 (SIRT1) activity. In non-tumor cells, several effects of abolished by inhibiting SIRT1, indicating mediation SIRT1. Melatonin is, addition to its antioxidant roles, an stimulatory agent. However, it can act as either pro- or anti-inflammatory regulator context-dependent way. stimulate release cytokines other mediators, but also, under different conditions, suppress inflammation-promoting processes such NO release, activation cyclooxygenase-2, inflammasome NLRP3, gasdermin D, toll-like receptor-4 mTOR signaling, cytokine SASP (senescence-associated secretory phenotype), amyloid-β toxicity. It also activates network, which SIRT1 activation, upregulation Nrf2 downregulation NF-κB, IL-4 IL-10 involved. A perhaps crucial action may be promotion macrophage microglia polarization favor phenotype M2. addition, many factors networks subject regulation microRNAs that target mRNAs respective upregulate them targeting their inhibitor proteins.

Aging is related to a number of functional and morphological changes leading progressive decline the biological functions an organism. Reactive Oxygen Species (ROS), released by several endogenous exogenous processes, may cause important oxidative damage DNA, proteins, lipids, cellular dysfunctions. The imbalance between ROS production antioxidant defenses brings stress conditions and, accumulation ROS, aging-associated diseases. purpose this review provide overview most relevant data reported in literature on natural compounds, mainly phytochemicals, with activity their potential protective effects age-related diseases such as metabolic syndrome, diabetes, cardiovascular disease, cancer, neurodegenerative chronic inflammation, possibly lower side effects, when compared other drugs.

The effect of immunometabolism on age-associated diseases remains uncertain. In this work, we show that T cells with dysfunctional mitochondria owing to mitochondrial transcription factor A (TFAM) deficiency act as accelerators senescence. mice, these instigate multiple aging-related features, including metabolic, cognitive, physical, and cardiovascular alterations, which together result in premature death. cell metabolic failure induces the accumulation circulating cytokines, resembles chronic inflammation is characteristic aging ("inflammaging"). This cytokine storm itself acts a systemic inducer Blocking tumor necrosis factor-α signaling or preventing senescence nicotinamide adenine dinucleotide precursors partially rescues mice

Low-grade inflammation is a hallmark of old age and central driver ageing-associated impairment disease1. Multiple factors can contribute to inflammation2; however, the molecular pathways that transduce aberrant inflammatory signalling their impact in natural ageing remain unclear. Here we show cGAS-STING pathway, which mediates immune sensing DNA3, critical chronic functional decline during ageing. Blockade STING suppresses phenotypes senescent human cells tissues, attenuates ageing-related multiple peripheral organs brain mice, leads an improvement tissue function. Focusing on brain, reveal activation triggers reactive microglial transcriptional states, neurodegeneration cognitive decline. Cytosolic DNA released from perturbed mitochondria elicits cGAS activity microglia, defining mechanism by engaged brain. Single-nucleus RNA-sequencing analysis microglia hippocampi gain-of-function mouse model demonstrates engagement sufficient direct states leading bystander cell inflammation, neurotoxicity impaired memory capacity. Our findings establish pathway as blockade potential strategy halt neurodegenerative processes age.

Inflammation is an integral part of animal biology. It provides critical protection from adverse environmental factors by enforcing the defense homeostasis and functional structural integrity tissues organs. Recent advances have uncovered a broad range biological processes that involve inflammatory control, calling for renewed framework inflammation beyond its classical roles in infection injury. In this Review, new perspectives on biology are discussed research directions suggested to address fundamental gaps our current understanding.

The unprecedented increase of life expectancy challenges society to protect the elderly from morbidity and mortality making vaccination a crucial mean safeguard this population. Indeed, infectious diseases, such as influenza pneumonia, are among top killers people in world. Elderly individuals more prone severe infections less responsive prevention, due immunosenescence combined with progressive proinflammatory status characteristic aging process (inflammaging). These factors responsible for most age-related diseases correlate poor response vaccination. Therefore, it is utmost interest deepen knowledge regarding role inflammaging responsiveness support development effective strategies designed elderly. In review we analyse impact age-associated inflammaging, immunobiography on immune elderly, consider systems biology approaches integrating multitude data order rationally design specifically tailored
