At the turn of century, researchers discovered a unique subtype T helper cells that secretes IL-17 and defined it as Th17. The latest study found Th17 play both positive negative definitive roles in regulation antitumor immune responses. Although function tumor microenvironment remains poorly understood, more studies have shown this paradoxical dual role is closely related to plasticity recent decades. Further understanding characteristics could yield novel useful therapeutic approaches treat cancer. In review, we further present high Th17-producing immunity.

Both T-cell deprivation and insufficient tumor immunogenicity seriously hinder the efficacy of immune-mediated destruction in melanoma. In this work, an amphiphilic polyethylene glycol-poly(2-hexoxy-2-oxo-1,3,2-dioxaphospholane) copolymer with a thermally sensitive flowable core (mPEG-b-PHEP) was chosen to incorporate IR780 dye manganese zinc sulfide nanoparticles (ZMS) form polymer micelles (denoted PPIR780-ZMS), which precisely controlled release ZMS after being triggered by near-infrared light (NIR). Mn2+-mediated chemodynamic therapy (CDT) photothermal trigger boosted generation reactive oxygen species (ROS), making PPIR780-ZMS smart bomblets vivo. It demonstrated that could maximize immunogenic cell death (ICD) cancer, is characterized abundant damage-associated molecular pattern (DAMP) exposure. As result, cytotoxic T cells (CD8+) helper (CD4+) expanded infiltrated neoplastic foci, further reprogrammed suppressive microenvironment (TME) against primary pulmonary metastases safe systemic cytokine expression. addition, cGAS-STING signaling pathway activation enhanced antitumor immunity nanocomposite, providing practical strategy for expanding use Mn-based nanostructures.

T lymphocytes (T cells) are divided into two functionally different subgroups the CD4+ helper cells (Th) and CD8+ cytotoxic (CTL). Adequate CD4 CD8 cell activation to proliferation, clonal expansion effector function is crucial for efficient clearance of infection by pathogens. Failure do so may lead exhaustion. Abstract Activation of the cyclic GMP‐AMP synthase‐stimulator interferon gene (cGAS‐STING) pathway is a potent anticancer immunotherapeutic strategy, and induction pyroptosis feasible way to stimulate immune responses. Herein, two Pt II complexes ( Pt1 Pt2 ) were designed as photoactivators cGAS‐STING pathway. Cyclooxygenases-2 (COX-2) and Prostaglandin E2 (PGE2), which are important in chronic inflammatory diseases, can increase tumor incidence promote growth metastasis. PGE2 binds to various prostaglandin E receptors activate specific downstream signaling pathways such as PKA pathway, β-catenin NF-κB pathway PI3K/AKT all of play roles biological pathological behavior. Nonsteroidal anti-inflammatory drugs (NSAIDs), COX-2 inhibitors, EP antagonists anti-tumor immune evasion. The COX-2-PGE2 promotes evasion by regulating myeloid-derived suppressor cells, lymphocytes (CD8+ T CD4+ cells natural killer cells), antigen presenting (macrophages dendritic cells). Based on conventional treatment, the addition inhibitors or may enhance immunotherapy response escape. However, there still a lot challenges cancer immunotherapy.
