DNA nanotechnology is a unique field, where physics, chemistry, biology, mathematics, engineering, and materials science can elegantly converge. Since the original proposal of Nadrian Seeman, significant advances have been achieved in past four decades. During this glory time, origami technique developed by Paul Rothemund further pushed field forward with vigorous momentum, fostering plethora concepts, models, methodologies, applications that were not thought before. This review focuses on recent progress origami-engineered nanomaterials five years, outlining exciting achievements as well unexplored research avenues. We believe spirit assets Seeman left for scientists will continue to bring interdisciplinary innovations useful next decade.

Protein-based virus-like particles (P-VLPs) are commonly used to spatially organize antigens and enhance humoral immunity through multivalent antigen display. However, P-VLPs thymus-dependent that themselves immunogenic can induce B cell responses may neutralize the platform. Here, we investigate thymus-independent DNA origami as an alternative material for display using receptor binding domain (RBD) of SARS-CoV-2 spike protein, primary target neutralizing antibody responses. Sequential immunization mice with DNA-based VLPs (DNA-VLPs) elicits protective antibodies in a manner depends on valency displayed T help. Importantly, immune sera do not contain boosted, class-switched against scaffold, contrast elicit strong memory both scaffold. Thus, DNA-VLPs immunogenicity without generating scaffold-directed thereby offer important particulate vaccine design.

The clustering of death receptors (DRs) at the membrane leads to apoptosis. With goal treating tumours, multivalent molecular tools that initiate this mechanism have been developed. Wireframe DNA origami can be used to fabricate virus-like particles for a range of biomedical applications, including the delivery nucleic acid therapeutics. However, acute toxicity and biodistribution these wireframe nanoparticles (NANPs) have not been previously characterized in animal models. In present study, we observed no indications BALB/c mice following therapeutically relevant dosage nonmodified DNA-based NANPs via intravenous administration, based on liver kidney histology, biochemistry, body weight. Further, immunotoxicity was minimal, as indicated by blood cell counts type-I interferon pro-inflammatory cytokines. an SJL/J model autoimmunity, NANP-mediated DNA-specific antibody response or immune-mediated pathology intraperitoneal administration NANPs. Abstract The natural micro‐ and nanoscale organization of biomacromolecules is a remarkable principle within living cells, allowing for the control cellular functions by compartmentalization, dimensional diffusion substrate channeling. In order to explore these biological mechanisms harness their potential applications such as sensing catalysis, molecular scaffolding has emerged promising approach. case synthetic enzyme cascades, developments in DNA nanotechnology have produced particularly powerful scaffolds whose addressability can be programmed with nanometer precision. this minireview, we summarize recent field biomimetic multicatalytic cascade reactions organized on nanostructures. We emphasize impact underlying design principles like origami, efficient strategies immobilization, well importance experimental parameters theoretical modeling. show how nanostructures enabled better understanding compartmentalization effects at length scale, discuss challenges future commercial applications.

Abstract DNA nanostructures with diverse biological functions have made significant advancements in biomedical applications. However, a universal strategy for the efficient production of is still lacking. In this work, facile and mild method presented self‐assembling polyethylenimine‐modified carbon dots (PEI‐CDs) into nanospheres called CANs at room temperature. This makes universally applicable to multiple applications involving various types DNA. Due ultra‐small size strong cationic charge PEI‐CDs, exhibit dense structure high loading capacity encapsulated while providing excellent stability by protecting from enzymatic hydrolysis. Additionally, Mg 2+ incorporated form Mg@CANs which enriches performance enables subsequent imaging exogenous . Especially, DNAzyme logic gate system that contains AND OR constructed successfully delivered tumor cells vitro vivo. They can be specifically activated endogenic human apurinic/apyrimidinic endonuclease 1 recognize expression levels miRNA‐21 miRNA‐155 sites biocomputing. A versatile pattern delivery flexible circuits miRNAs are developed.
