A novel SNP of TECPR2 gene associated with heat tolerance in Chinese cattle DOI

Heat stress affects the animal production and causes serious economic losses to husbandry. Tectonin beta-propeller repeat containing 2 (TECPR2) gene plays an important role in autophagy which may affect temperature sensation animals. A missense mutation (XM_024981840.1:c.3989 G > p.Arg1330His) of transcripts X4 bovine TECPR2 was identified. In this study, c.3989 variant genotyped a total 25 cattle breeds (520 individuals). Our results indicated that frequency allele showed decreasing pattern from southern northern cattle, while opposite pattern, consistent with climate distribution China. Compared GG genotype, carried more AA AG genotypes. Furthermore, association out frequencies genotypes (GG, AG, AA) value parameters (mean annual (T), relative humidity (RH) index (THI) were significantly correlated (p < 0.01). Hence, we speculated associated heat tolerance trait Chinese locus be considered as molecular marker for breeding.

Heat stress is the major factor affecting cattle fertility but molecular mechanisms of deleterious impacts elevated temperature on oocyte are still not well known. Therefore, aim this study to gain a better understanding underlying mechanism how heat affects GV-stage and MII-stage oocytes discover hub genes resistance for cow oocytes. In study, we used bioinformatics approach differentially expressed between oocytes, which were collected during spring summer. When compared in July (Jul DEGs group) total 1068 found as result stress. Also, HSPA8, COPS5, POLR2L, PSMC6, TPI1 identified common top ranked Jul group. The highest connected gene group was determined HSPA8. Our results showed that different response might be activated protect from temperatures cattle. their associated pathways play an important role

High summer temperatures have deleterious effects on oocyte developmental competence. The antioxidant and autophagy-related properties of 5-aminolevulinic acid (5-ALA) gives the compound a broad range biological activities. This study aimed to evaluate of: 1) high temperature-humidity index (THI) competence bovine oocytes, 2) 5-ALA administration in combination with sodium ferrous citrate (SFC) during vitro maturation (IVM) evaluated at THI. Bovine ovaries were collected from local slaughterhouse moderate environmental temperature (MT; THI 56.2) (HT; 76.7) periods; cumulus-oocyte complexes (COCs) aspirated medium-sized follicles, matured for 22 h, fertilized, cultured 10 days. For COCs HT period, 0 (control), 0.01, 0.1, 0.5, or 1 µM was added medium SFC molar ratio 1:0.125. results showed that adversely affected blastocyst hatching rates compared MT. Adding 5-ALA/SFC (1 µM/0.125 µM) oocytes period improved cumulus cell expansion no-addition control. In conclusion, this can disrupt ameliorates negative effect heat stress improves subsequent embryo development.

A high temperature-humidity index during summer has deleterious effects on mitochondrial function, reducing oocyte developmental competence. 5-Aminolevulinic acid (5-ALA) and sodium ferrous citrate (SFC) are both known to support function have strong anti-oxidant anti-apoptotic activities. This study aimed determine the mechanism of action 5-ALA/SFC quality. Bovine oocytes were collected from medium-sized follicles (July-September, index:76.6), cultured with 0, 1, 2, 4, 8 µM 5-ALA SFC at a molar ratio 1:0.125, fertilized, for 10 days. The addition 8/1 had effect cleavage rate in comparison control oocytes, but did not affect blastocyst rate, while 1/0.125 significantly higher increase than 5-ALA/SFC. 2/0.25 improved quality by increasing distribution pattern metaphase-II reactive oxygen species upregulating nuclear factor erythroid-2-related heme oxygenase-1, superoxide dismutase-1 2 transcription cumulus cells. These results indicate that may competence provide actions cumulus-oocyte complexes.

The transition from late pregnancy (LP) to early lactation (EL) in dairy cows is characterized by a major reorganization of the metabolic activities liver and adipose tissue support milk synthesis.
