The COVID-19 pandemic ignited research centered around the identification of robust biomarkers and therapeutic targets. SARS-CoV-2, virus responsible, hijacks metabolic machinery host cells. It relies on lipids lipoproteins cells for entry, trafficking, immune evasion, viral replication, exocytosis. infection causes cell lipid remodelling. Targeting lipid-based processes is thus a promising strategy countering COVID-19. Here, we review role in different steps SARS-CoV-2 pathogenesis identify lipid-centric targetable avenues. We discuss lipidome changes infected patients their relevance as potential clinical diagnostic or prognostic biomarkers. summarize emerging direct indirect approaches targeting using lipid-inspired approaches. Given that protein-targeted therapies may become less effective due to mutations variants, interventions provide additional perhaps better means combating this future pandemics.

In this research, a bioinformatics analysis was performed to carry out physicochemical properties biological activities phytochemicals in analyze binding activities, targets, functions UC. This shows that CR’s key phytochemicals, which are named Coptisine, Berberrubine, Berlambine, Berberine, Epiberberine, Obacunone, Worenine, Quercetin, (R)-Canadine, Magnograndiolide, Palmatine Moupinamide, have ideal bioactivity. A total 1,904 potential phytochemical 17,995 UC-related identified, we finally acquire 233 intersection between disease. protein-protein interaction network common constructed; six hub were acquired with degree greater than or equal median, namely TP53, HSP90AA1, STAT3, ESR1, MYC, RELA. The enrichment suggested core may exert an impact on anti-inflammatory, immunoregulatory, anti-oxidant anti-fibrosis mainly through PI3K/ART signaling pathway, Th17 differentiation inflammatory bowel disease etcetera. Also, docking strong affinity targets. Finally, CR, GO functions, KEGG pathways constructed. study indicates superior drug likeness bioactivity, mechanism be via pathway mentioned above. critical pharmacological provide direction future research.

Pregnant women infected with SARS-CoV-2 in early pregnancy may face an increased risk of miscarriage due to immune imbalance at the maternal-fetal interface. However, molecular mechanisms underlying crosstalk between COVID-19 infection and recurrent spontaneous abortion (RSA) remain poorly understood. This study aimed elucidate transcriptomic dialog RSA. Based on bioinformatics analysis, 307 common differentially expressed genes were found (GSE171110) RSA (GSE165004). Common DEGs mainly enriched ribosome-related cell cycle-related signaling pathways. Using degree algorithm, top 10 hub (RPS27A, RPL5, RPS8, RPL4, RPS2, RPL30, RPL23A, RPL31, RPL26, RPL37A) selected from based their scores. The results qPCR general agreement raw letter analysis. candidate drugs also P-values. In this study, we provide markers, pathways, small molecule compounds that associate COVID-19. Introduction The key to understanding the COVID-19 correlates of protection is assessing vaccine-induced immunity in different demographic groups. Young people are at a lower risk mortality, females than males, and often generate stronger immune responses vaccination. Methods We studied two doses BNT162b2 Pfizer vaccine an adolescent cohort (n = 34, ages 12–16), age group previously shown elicit significantly greater same young adults. Adolescents were with aim comparing their response that adults; assess impacts other factors such as sex, ongoing SARS–CoV–2 infection schools, prior exposure endemic coronaviruses circulate high levels people. At time, we able evaluate co-administered live attenuated influenza vaccine. Blood samples from 34 adolescents taken before after vaccination vaccines assayed for SARS–CoV–2-specific IgG neutralising antibodies cellular specific betacoronaviruses. targeting lineages contained also assessed. Results Robust identified infected one dose, required infection-naïve adolescents. As demonstrated, total SARS–CoV-2 Spike higher among vaccinated adults (aged 32–52) who received (comparing infection-naïve, 49,696 vs. 33,339; p 0.03; SARS-CoV–2 infected, 743,691 269,985; <0.0001) by MSD v-plex assay. There was no evidence compared males. Discussion These findings may result introduction novel mRNA platforms, generating patterns divergent established trends providing new insights into what might be protective following

Patients with lung adenocarcinoma (LUAD) are vulnerable to COVID-19. However, there is currently a lack of available treatments for LUAD patients infected Jaranol naturally derived flavonoid that exhibits promising properties as an antiviral and antitumor agent. Therefore, this research intends investigate the molecular targets underlying mechanisms jaranol treatment COVID-19/LUAD. Through network pharmacology, 47 intersection target genes were identified against COVID-19/LUAD, eight-gene risk score model strong predictive accuracy was constructed. Functional enrichment analysis demonstrated main mechanism in combating COVID-19/LUAD involved regulation response hormone, positive phosphorylation, PI3K-Akt signaling pathway. Furthermore, results docking indicated exhibited significant affinity eight hub (AKT1, SRC, EGFR, HSP90AA1, ESR1, PTGS2, PPARG, MMP9) well three core COVID-19-related (ACE2, 3CLpro, PLpro). The direct binding between MMP9 then validated A549 cancer cells using cellular thermal shift assay (CETSA). These findings suggest has therapeutic effect providing theoretical foundation novel perspectives future development potential pharmaceutical candidate.

Abstract The risk of Alzheimer’s disease (AD) in women is about 2 times greater than men. estrogen hypothesis being accepted as the essential sex factor causing difference AD. Also, recent meta-analysis using large-scale medical records data indicated replacement therapy. However, underlying molecular targets and mechanisms explaining this AD development remain unclear. In study, we identified that treatment can strongly inhibition neuro-inflammation signaling targets, systems pharmacology model; ESR1/ESR2 (the receptors estrogen) are topologically close to neuroinflammation biomarker genes network analysis. Moreover, level decreased an extremely lower men after age 55. Pooling together multiple pieces evidence, it concluded loss unleashing increases women’s disease. These analysis results provide novel supporting evidence potential mechanism anti-neuroinflammation role Medications boosting direct downstream ESR1/ESR2, or inhibiting upstream neuroinflammation, like JAK2 inhibitors, on be potentially effective synergistic combined with for prevention treatment.
