Fatty acid synthase (FAS) is a key enzyme in the lipid synthesis pathway, however, its roles insects remain largely unknown. Here, we firstly identified two FAS genes from transcriptome dataset of general cutworm Spodoptera litura, which destructive insect pest many crops. Both SlFAS1 and SlFAS2 were highly expressed third instar larvae their fat bodies. Then, successfully silenced content α-linolenic triglyceride was significantly decreased. Besides that, effect on metamorphic development S. litura evaluated. The results indicate that after silencing SlFAS1, survival rates decreased compared to control groups. Silencing fifth resulted more malformed pupae adults, emergence reduced. Furthermore, ecdysone haemolymph SlFAS1. In addition, knocking down alters expression other lipogenesis implying has an impact pathway. present study deepens understanding provides novel potential targets for managing pests.

Abstract Fatty acid synthase is a multifunctional enzyme involved in the formation of fatty acids. Despite role acids cell signalling and energy metabolism, as precursors to pheromones hydrocarbons that waterproof cuticle, insect synthases have been scarcely studied. Here we perform molecular characterization three genes ( RPRC000123 , RPRC000269 RPRC002909 ) Chagas disease vector, Rhodnius prolixus . Gene expression screening by reverse transcription quantitative PCR showed are expressed almost exclusively integument tissue whilst mostly fat body also several organs. Phylogenetic analysis, together with gene results, orthologues Drosophila melanogaster 1 FASN1 ), FASN2 FASN3 genes, respectively. After RNA interference‐mediated knockdown insects died immediately after moulting next developmental stage. However, mortality was prevented placing under saturated humidity conditions, suggesting dehydration might play insects’ death. Lipid analyses ‐silenced reduced amounts methyl‐branched hydrocarbons, compared controls. These data support an important for biosynthesis cuticle.

Juvenile hormones (JH) and ecdysteroids control postembryonic development in insects. They serve as valuable targets for pest management. Hence, understanding the molecular mechanisms of their action is crucial importance. CREB-binding protein (CBP) a universal transcriptional co-regulator. It controls expression several genes including those from hormone signaling pathways through co-activation many transcription factors. However, role CBP during insects not well understood. Therefore, we have studied Tribolium, model coleopteran insect. ubiquitously expressed red flour beetle, Tribolium castaneum. RNA interference (RNAi) mediated knockdown resulted decrease JH induction Kr-h1 gene larvae led to block development. Moreover, injection double-stranded (dsRNA) showed lethal phenotypes within 8 days injection. RNA-seq subsequent differential analysis identified target Tribolium. Knockdown caused 1306 coding factors other proteins associated with growth Depletion impaired response (e.g., Kr-h1, Hairy, early trypsin) ecdysone (EcR, E74, E75, broad complex). Further, GO enrichment analyses downregulated different functions developmental processes, pigmentation, anatomical structure development, regulation biological cellular etc. These data suggest diverse but roles insect, can RNAi management this stored product pest.

Real-time PCR (RT-qPCR) expression analysis is a powerful analytical technique, but for normalization of data requires the use stable reference genes. However, suitable genes are still not known in case Phenacoccus solenopsis under variable experimental treatments. The present study focused on identification housekeeping as different abiotic and biotic factors P. solenopsis. We analyzed relative six commonly used candidate developmental stages, host-feeding assay, temperature treatments field distribution conditions. Expression stabilities were by geNorm, NormFinder, RefFinder. Under conditions, β-Tubulin was found to be most followed rpl32 α-Tubulin. In host feeding treatment α-tubulin identified genes, while stress, combination α-Tubulin normalizing RT-qPCR data. Further, above-identified validated using based gene four objective namely, Myoinhibitory peptides (MIPs), Zinc_metalloprotease (Zn_Mp), fatty acid synthase (fas) alpha-glucosidase. Abstract Lipids of the insect cuticle have important roles in resistance against arid environment and invasion foreign substances. Fatty acid synthase (FAS) is an enzyme lipid synthesis pathway. In present study, we identified three FAS genes from transcriptome data migratory locust, Locusta migratoria , based on bioinformatics analyses. Among them, two ( LmFAS1 LmFAS3 ) are highly expressed integument fifth instar nymphs. Suppression by RNA interference caused lethality during ecdysis or shortly after moulting. The weight locusts content droplets were reduced compared with those control. results gas chromatography–mass spectrometry analysis showed that knockdown led to a decrease both cuticular hydrocarbons inner (CHCs IHCs) contents, especially methyl branched hydrocarbons. By contrast, only resulted IHC content, but not CHCs. consequence, LmFAS1‐ ‐suppressed locusts, hydrocarbon deficiency desiccation enhanced permeability sensitivity insecticides. These indicate essential for production permeability, which play influential waterproofing cuticle.
