Toll-like receptors (TLRs) are a well-known family of pattern recognition that play an important role in host immune system. TLR triggering leads to the induction pro-inflammatory cytokines and chemokines, driving activation both innate adaptive immunity. Recently, increasing number studies have shown link between TLRs cancer. Among them, toll-like receptor 4 (TLR4) signaling pathway is associated with inflammatory response cancer progression. Dietary phytochemicals potential modulators immunological status various pharmacological properties including anti-cancer, anti-oxidant anti-inflammatory. The liposomal delivery system drawn attention as one the noteworthy approaches to increase dissolution and subsequently absorption gastrointestinal (GI) tract because its biocompatibility ability encapsulate hydrophobic molecules lipid domain. However, there have several drawbacks, such structural instability GI permeability across intestinal epithelia relatively large size. In addition, no formulations approved for oral date, despite success parenteral liposomes. Nevertheless, resurged rapid published studies last decade. it is discouraging most this research vitro only not many vivo data. present review focused on evidence improved BA using liposomes resolve doubts raised concerning attempted provide insight by highlighting used achievements.

Curcumin, a lipophilic polyphenol contained in the rhizome of Curcuma longa (turmeric), has been used for centuries traditional Asian medicine, and nowadays it is widely food as dietary spice worldwide. It received considerable attention its pharmacological activities, which appear to act primarily through anti-inflammatory antioxidant mechanisms. For this reason, proposed tool management many diseases, among are gastrointestinal neurological diabetes, several types cancer. However, pharmacology curcumin remains be elucidated; indeed, discrepancy exists between well-documented vitro vivo activities poor bioavailability chemical instability that should limit any therapeutic effect. Recently, hypothesized could exert direct regulative effects tract, where high concentrations have detected after oral administration. Consequently, might directly exerts regulatory on gut microbiota, thus explaining paradox low systemic wide activities. well known microbiota important roles human physiology, composition can influenced by multitude environmental lifestyle factors. Accordingly, perturbations microbiome profile or dysbiosis key role disease progression. Interestingly, metabolites shown influence microbiota. worth noting from interaction two different phenomena arise: regulation intestinal microflora biotransformation both them potentially crucial activity. Curcumin possesses a plethora of interesting pharmacological effects. Unfortunately, it is also characterized by problematic drug delivery and scarce bioavailability, representing the main problem related to use this compound. Poor absorption, fast metabolism, rapid systemic clearance are most important factors contributing low curcumin levels in plasma tissues. Accordingly, overcome these issues, numerous strategies have been proposed investigated article. Due advances field, we describe here promising for increasing including adjuvant, complexed/encapsulated curcumin, specific formulations, nanoparticles. We analyze current strategies, already available market, advanced technologies that can offer future perspective effective formulations. focus attention on effectiveness curcumin-based formulations clinical trials, providing comprehensive summary. Clinical trial results, employing various methods showed improved bioavailability corresponds increased therapeutic efficacy. Furthermore, field nanoparticles hold great promise developing complexes as agents. Summarizing, suitable polyphenol will ensure possibility using curcumin-derived practice preventive disease-modifying therapeutics.
