Radiotherapy is widely applied for multiple malignant tumors ablation in the clinic. However, redundant doses of X-rays might destroy normal tissue periphery tumor sites. Here, we developed an integrated nanosystem (Bac@BNP) composed engineered bacteria (Bac) and Bi2S3 nanoparticles (BNPs) sensitizing radiotherapy. Bac could target colonize sites alternatively, which overexpressed cytolysin A (ClyA) protein to regulate cell cycle from a radioresistant phase radiosensitive phase. Simultaneously, peptide-modified BNPs, as radiosensitizer with high-Z element, was released surface owing matrix metalloproteinase-2 (MMP-2) response microenvironment. Under X-ray irradiation, BNPs enhance radiotherapy sensitivity by triggering intracellular generation reactive oxygen species (ROS), coupled DNA damage. In this constructed nanosystem, combination Bac@BNP irradiation led significant suppression breast carcinoma murine models reduced side effects.

Radiation detection, using materials to convert high-energy photons low-energy (X-ray imaging) or electrical charges detector), has become essential for a wide range of applications including medical diagnostic technologies, computed tomography, quality inspection and security, etc. Metal halide perovskite-based high-resolution scintillation-imaging screens direct conversion detectors are promising candidates such applications, because they have high absorption cross sections X-rays due their heavy atom (e.g., Pb2+, Bi3+, I–) compositions; moreover, these solution-processable at low temperature, possessing tunable bandgaps, near-unity photoluminescence quantum yields, trap density, charge carrier mobility, fast photoresponse. In this review, we explore decipher the working mechanism scintillators as well key advantages perovskites both detection approaches. We further discuss recent advancements in research area, pointing out remaining challenges our perspective future directions toward X-ray applications.

Abstract Having a hypoxic microenvironment is common and salient feature of most solid tumors. Hypoxia has profound effect on the biological behavior malignant phenotype cancer cells, mediates effects chemotherapy, radiotherapy, immunotherapy through complex mechanisms, closely associated with poor prognosis in various patients. Accumulating studies have demonstrated that normalization tumor vasculature, nanoparticle carriers biocarriers can effectively increase oxygen concentration microenvironment, improve drug delivery efficacy radiotherapy. They also infiltration innate adaptive anti-tumor immune cells to enhance immunotherapy. Furthermore, drugs targeting key genes hypoxia, including hypoxia tracers, hypoxia-activated prodrugs, hypoxia-inducible factors downstream targets, be used for visualization quantitative analysis antitumor activity. However, relationship between an area research requires further exploration. Nanotheranostics is one of the biggest scientific breakthroughs in nanomedicine. Most currently available diagnosis and therapies are invasive, time-consuming, associated with severe toxic side effects. Nanotheranostics, on other hand, has potential to bridge this gap by harnessing capabilities nanotechnology nanomaterials for combined therapeutics diagnostics markedly enhanced efficacy. Janus nanoparticles (JNPs) refer to the integration of two or more chemically discrepant composites into one structure system. Studies JNPs have been significant interest due their interesting characteristics stemming from asymmetric structures, which can integrate different functional properties and perform synergetic functions simultaneously. Herein, we present recent progress particles, comprehensively detailing fabrication strategies applications. First, classification is divided three blocks, consisting polymeric composites, inorganic hybrid polymeric/inorganic composites. Then, are alternately summarized, examining self-assembly strategy, phase separation seed-mediated polymerization, microfluidic preparation nucleation growth methods, masking methods. Finally, various intriguing applications presented, including solid surfactants agents, micro/nanomotors, biomedical such as biosensing, controlled drug delivery, bioimaging, cancer therapy, combined theranostics. Cancer immunotherapy has achieved promising clinical progress over the recent years for its potential to treat metastatic tumors and inhibit their recurrences effectively. Chemotherapy causes off-target toxicity and is often ineffective against solid tumors. Targeted on-demand release of chemotherapeutics remains a challenge. Here, cancer-cell-membrane-coated mesoporous organosilica nanoparticles (MONs) containing X-ray- reactive oxygen species (ROS)-responsive diselenide bonds for controlled doxorubicin (DOX) at tumor sites are developed. DOX-loaded MONs coated with 4T1 breast cancer cell membranes (CM@MON@DOX) show greater accumulation prolonged blood circulation time versus an uncoated control in mice bearing orthotopic mammary Under low-dose X-ray radiation, the exhibit carrier degradation-controlled via cleavage bonds, resulting DOX-mediated immunogenic death site. Combination PD-L1 checkpoint blockade further enhances inhibition growth metastasis low systemic toxicity. Together, findings promise these biomimetic, radiation-responsive diselenide-bond-bridged chemo-immunotherapy.
