The present study aims to evaluate the effects of antibacterial peptide Microcin J25 (ABP MccJ25) on growth performance, intestinal barrier function, antioxidant capacity and ileal microbiota in pigeon squabs. 80 pairs American Silver King parents 240 healthy day 1 squabs were randomly divided into four groups, including 4 parent 12 per group with 5 replicates. Parent pigeons fed a basal diet (CON) or addition 100, 200, 300 mg/kg ABP MccJ25 (ABP100, ABP200, ABP300, respectively) for weeks. Results showed that ABP200 remarkably reduced feed conversion ratio increased average daily gain from 7 21 d (p < 0.05), improved serum 14 28 0.05). Villus height villus crypt depth duodenum, jejunum, ileum significantly groups ABP300 In addition, obviously up-regulated relative mRNA expression antioxidant-related genes barrier-related Moreover, proportion beneficial bacteria Firmicutes Lactobacillales, which favourable health noticeably Taken together, these results indicated enhanced capacity, strengthened facilitated regulation microbiota, leading improvement performance Therefore, this provides support application 200 production.

Although leaky gut syndrome is not recognized as an official diagnosis for human diseases, it now believed that dysfunction of the cell barrier causes increased permeability intestinal epithelial cells leading to this condition. Probiotics have been widely used improve health, and studies investigated relevance protecting by taking probiotic strains in vitro vivo. However, most restricted use single or several do consider commercially available products composed multi-species. In study, we provide experimental evidence a multi-species mixture eight different heat-treated strain effective preventing conditions. We employed co-culture model system utilizing two differentiated lines mimic tissue. The integrity function was protected preserving occludin protein level activating AMPK signaling pathway, associated with tight junctions (TJs), through treatment Caco-2 cells. Moreover, confirmed application reduced expression proinflammatory cytokine genes inhibiting NFκB pathway when artificial inflammation induced system. Finally, proved measured trans-epithelial electrical resistance (TEER) significantly decreased treated cells, indicating compromised. exhibited protective effect on via enhancing TJ complexes reducing inflammatory responses

Changes in the functions of intestinal barrier occur parallel with development sepsis. The protection by Bifidobacterium strains (BB, BL, BB12, and BLBB) was evaluated mice injected lipopolysaccharide (LPS). results revealed an increase ratio ileal villus length to crypt depth BLBB group compared that LPS group, as were number IgA+ plasma, CD4+/CD8+ T, dendritic cells. levels diamine oxidase (DAO) d-lactic acid serum lessened after injection group. In addition, exhibited increased expression level tight junction proteins (zonula occludens-1, occludin, claudin-1), mucin (MUC2) mRNA, reduced NFκB/MAPK signaling pathways, decreased inflammatory cytokines (IL-1β, IL-6, TNF-α). enriched relative abundance Muribaculaceae, Lachnospiraceae_NK4A136_group, Clostridia_Ucg-014, Alistipes, resulting producing short-chain fatty acids. Furthermore, leads higher deoxycholic biosynthesized linoleate. This study suggested could enhance capacity mucosal immunity, reduce inflammation, improve composition gut microbiota. bifidum E3 combined longum subsp. infantis E4 may thus serve a probiotic against injury caused LPS.

Inflammatory bowel disease (IBD) is a chronic inflammatory characterized by intestinal barrier dysfunction, synergistic effects and excessive tissue injury. Gallic acid (GA) renowned for its remarkable biological activity, encompassing anti-inflammatory antioxidant properties. However, the underlying mechanisms which GA protects against inflammation have not been fully elucidated. The aim of this study to investigate effect on lipopolysaccharide (LPS)-stimulated human colon carcinoma cell line (Caco-2) explore molecular mechanism involved. Our findings demonstrate that 5 μg/mL restores downregulation mRNA protein levels Claudin-1, Occludin, ZO-1 decreases expressions factors such as IL-6, IL-1β TNF-α induced LPS. In addition, exhibits protective reducing LPS-enhanced early late apoptotic ratios, downregulating pro-apoptotic (

Essential oils (EO) are known for their antioxidant, anti-inflammatory, antimicrobial, and growth-promoting properties. However, data rgarding impact on the intestinal health gut microbiota of ducks remain limited. Thus, this study aimed to investigate effects plant EO growth performance, health, Muscovy ducks. A total 360 healthy male aged 1 d were randomly divided into 4 groups with 6 replicates 15 per replicate. Ducks fed basal diets supplemented 0, 100, 200, or 300 mg/kg EO. The results showed that 200 supplementation significantly (P < 0.05) increased final body weight average daily gain, while decreased feed conversion ratio during 56-d experimental period. Furthermore, dietary enhanced antioxidant capacity immune function improved barrier intestine. Additionally, 16S rDNA sequencing analysis favorably modulated cecal microbial diversities composition evidenced by abundances short-chain fatty acid-producing bacteria (e.g., Subdoligranulum Shuttleworthia) potential enteric pathogenic Alistipes, Eisenbergiella, Olsenella). relative abundance beneficial was positively correlated immune, biomarkers. Overall, these findings revealed had several potentially performance improving capacity, enhancing modulating microbiota.

The impact of leptin resistance on intestinal mucosal barrier integrity, appetite regulation, and hepatic lipid metabolism through the microbiota–gut–brain–liver axis has yet to be determined. Water extract Phyllanthus emblica L. fruit (WEPE) its bioactive compound gallic acid (GA) effectively alleviated methylglyoxal (MG)-triggered in vitro. Therefore, this study investigated how WEPE GA intervention relieve resistance-associated dysfunction mucosa, appetite, accumulation high-fat diet (HFD)-fed rats. results showed that significantly reduced tissues (jejunum, brain, liver) MG-evoked resistance, malondialdehyde (MDA), proinflammatory cytokines, SOCS3, orexigenic neuropeptides, increasing receptor, tight junction proteins, antimicrobial peptides, anorexigenic excretion fecal triglyceride (TG), short-chain fatty acids (SCFAs) via a positive correlation with Allobaculum Bifidobacterium microbiota. These novel findings suggest holds potential as functional food ingredient for alleviating obesity complications.

The intestinal barrier dysfunction associated with chronic inflammation is a major health concern. This work aimed to investigate the protective effect and molecular mechanism of Senegalia macrostachya seed polysaccharide fraction (SMSP2) on inflammation-induced using Caco-2 cells RAW264.7 macrophage co-culture model. results showed that LPS stimulation basolateral compromised integrity apical differentiated monolayer, resulting in decreased transepithelial electrical resistance (TER) increased inflammatory markers. SMSP2 treatment maintained higher TER value reduced epithelial permeability lucifer yellow (LY) dye. In addition, group significant decrease mRNA level factors such as inducible nitric oxide synthase (iNOS), cyclooxygenase 2 (COX-2), interleukin (IL)-8, tumor necrosis factor-α (TNF-α) monolayer compared group. Moreover, expression tight junctions (TJ) zonula occludens (ZO-1), occludin, claudin-1 at protein levels. Furthermore, immunofluorescence assay ameliorated overall distribution TJ proteins monolayer. application also resulted downregulation nuclear factor kappa-B (NF-κB) phosphorylation myosin light chain kinase (MLCK) expression, which implies preserved from disruption through inhibition NF-kB-mediated MLCK signaling pathway activation. seeds could therefore be promising functional food used improve function.
