Changes in the functions of intestinal barrier occur parallel with development sepsis. The protection by Bifidobacterium strains (BB, BL, BB12, and BLBB) was evaluated mice injected lipopolysaccharide (LPS). results revealed an increase ratio ileal villus length to crypt depth BLBB group compared that LPS group, as were number IgA+ plasma, CD4+/CD8+ T, dendritic cells. levels diamine oxidase (DAO) d-lactic acid serum lessened after injection group. In addition, exhibited increased expression level tight junction proteins (zonula occludens-1, occludin, claudin-1), mucin (MUC2) mRNA, reduced NFκB/MAPK signaling pathways, decreased inflammatory cytokines (IL-1β, IL-6, TNF-α). enriched relative abundance Muribaculaceae, Lachnospiraceae_NK4A136_group, Clostridia_Ucg-014, Alistipes, resulting producing short-chain fatty acids. Furthermore, leads higher deoxycholic biosynthesized linoleate. This study suggested could enhance capacity mucosal immunity, reduce inflammation, improve composition gut microbiota. bifidum E3 combined longum subsp. infantis E4 may thus serve a probiotic against injury caused LPS.

However, most restricted use single or several do consider commercially available products composed multi-species. In study, we provide experimental evidence a multi-species mixture eight different heat-treated strain effective preventing conditions. We employed co-culture model system utilizing two differentiated lines mimic tissue. The integrity function was protected preserving occludin protein level activating AMPK signaling pathway, associated with tight junctions (TJs), through treatment Caco-2 cells. Moreover, confirmed application reduced expression proinflammatory cytokine genes inhibiting NFκB pathway when artificial inflammation induced system. Essential oils (EO) are known for their antioxidant, anti-inflammatory, antimicrobial, and growth-promoting properties. However, data rgarding impact on the intestinal health gut microbiota of ducks remain limited. Thus, this study aimed to investigate effects plant EO growth performance, health, Muscovy ducks. A total 360 healthy male aged 1 d were randomly divided into 4 groups with 6 replicates 15 per replicate. Ducks fed basal diets supplemented 0, 100, 200, or 300 mg/kg EO. The results showed that 200 supplementation significantly (P < 0.05) increased final body weight average daily gain, while decreased feed conversion ratio during 56-d experimental period. Furthermore, dietary enhanced antioxidant capacity immune function improved barrier intestine. Additionally, 16S rDNA sequencing analysis favorably modulated cecal microbial diversities composition evidenced by abundances short-chain fatty acid-producing bacteria (e.g., Subdoligranulum Shuttleworthia) potential enteric pathogenic Alistipes, Eisenbergiella, Olsenella). relative abundance beneficial was positively correlated immune, biomarkers. This work aimed to investigate the protective effect and molecular mechanism of Senegalia macrostachya seed polysaccharide fraction (SMSP2) on inflammation-induced using Caco-2 cells RAW264.7 macrophage co-culture model. results showed that LPS stimulation basolateral compromised integrity apical differentiated monolayer, resulting in decreased transepithelial electrical resistance (TER) increased inflammatory markers. SMSP2 treatment maintained higher TER value reduced epithelial permeability lucifer yellow (LY) dye. In addition, group significant decrease mRNA level factors such as inducible nitric oxide synthase (iNOS), cyclooxygenase 2 (COX-2), interleukin (IL)-8, tumor necrosis factor-α (TNF-α) monolayer compared group. Moreover, expression tight junctions (TJ) zonula occludens (ZO-1), occludin, claudin-1 at protein levels. Furthermore, immunofluorescence assay ameliorated overall distribution TJ proteins monolayer. application also resulted downregulation nuclear factor kappa-B (NF-κB) phosphorylation myosin light chain kinase (MLCK) expression, which implies preserved from disruption through inhibition NF-kB-mediated MLCK signaling pathway activation. seeds could therefore be promising functional food used improve function.
