Background Transcranial focused ultrasound (TFUS) is an emerging neuromodulation tool for temporarily altering brain activity and probing network functioning. The effects of TFUS on the default mode (DMN) are unknown. Objective study examined transcranial functional connectivity (DMN), specifically by targeting posterior cingulate cortex (PCC). Additionally, we investigated subjective mood, mindfulness, self-related processing. Methods employed a randomized, single-blind design involving 30 healthy subjects. Participants were randomly assigned to either active group or sham group. Resting-state magnetic resonance imaging (rs-fMRI) scans conducted before after application. Abstract Psychedelics offer a profound window into the human brain through their robust effects on perception, subjective experience, and activity patterns. The serotonergic psychedelic N,N-dimethyltryptamine (DMT) induces profoundly immersive altered state of consciousness lasting under 20 min, allowing entire experience to be captured during single functional magnetic resonance imaging (fMRI) scan. Abstract Modifying behaviors, such as alcohol consumption, is difficult. Creating psychological distance between unhealthy triggers and one’s present experience can encourage change. Using two multisite, randomized experiments, we examine whether theory-driven strategies to create distance—mindfulness perspective-taking—can change drinking behaviors among young adults without dependence via a 28-day smartphone intervention (Study 1, N = 108 participants, 5492 observations; Study 2, 218 9994 observations). 2 presents close replication with fully remote delivery during the COVID-19 pandemic.
