Abstract The potential of sponge-derived chemicals for pharmaceutical applications remains largely unexploited due to limited available biomass. Although many have attempted culture marine sponge cells in vitro create a scalable production platform such biopharmaceuticals, these efforts been mostly unsuccessful. We recently showed that Geodia barretti could divide rapidly M1 medium. In this study we established the first continuous cell line, originating from G. . cultured OpM1 medium, modification M1, grew more and higher density than M1. Cells reached 1.74 population doublings after 30 min, twofold already rapid growth rate 0.74 min maximum number increased 5 at least 98 OpM1. Subcultured be cryopreserved used inoculate new cultures. Abstract Abnormal activation of the nuclear factor‐kappa B (NF‐κB) signaling pathway is closely implicated in triple‐negative breast cancer growth, metastasis, and tumor immune escape. In this study, anti‐cancer effects icariin, a natural flavonol glycoside, toward cells underlying mechanisms were investigated. This investigation showed that icariin selectively inhibited proliferation triggered apoptosis concentration‐ time‐dependent manner, but exhibited little cytotoxicity normal cells. Moreover, induced cell via mitochondria‐mediated pathway, as indicated by upregulated ratio Bax/Bcl‐2 reactive oxygen species induction. Natural products (NPs) or their derivatives represent a large proportion of drugs that successfully progress through clinical trials to approval. This study explores the presence NPs in both early- and late-stage drug discovery determine success rate, factors features natural contribute such success. As proxy for early development stages, we analyzed patent applications over several decades, finding consistent NP, NP-derived, synthetic-compound-based documents, with latter group outnumbering NP NP-derived ones (approximately 77% vs 23%). We next assessed trial data, where observed steady increase compounds from phases I III (from approximately 35% phase 45% III), an inverse trend synthetics 65% 55% III). Finally, vitro silico toxicity studies revealed were less toxic alternatives synthetic counterparts. These discoveries offer valuable insights successful NP-based development, highlighting potential benefits prioritizing as starting points.

Since the early research efforts focusing on bioactive marine substances such as spongouridine, from sponge Cryptotethya crypta, natural products (MNPs) have arisen a robust and sustainable supplier for drug leads. Marine present definite, unprecedented structural diversifications varieties of interesting biomedical potentialities with novel mechanisms action. Until today, eight clinically approved products-based drugs by two worldwide medical organizations (including U.S. FDA, European Medicines Agency (EMEA)), been developed treatment different forms carcinoma, pain, Alzheimer's disease other current challenges. Recent clinical trial analysis disclosed that pipeline contains more than twenty listed candidates in trials phase III, II, or I. Herein, we recent insights centered to preclinically investigated compounds, well sampling techniques, extraction & identification tools, classification MNPs, some reported biological activities, challenges faced during MNPs development.
