Abstract Sex determination is a variable and complex mechanism, yet it can be found all over the plant animal kingdoms. It creates two morphological different outcomes from one same species. Our work demonstrates involvement of non-coding RNA dnm3os , its embedded microRNA miR214 in this process for teleost Danio rerio . First, we find that acts through gsdf to drive female development. Second, additional alleles revealed lncRNA also promote male development unknown mechanisms. The genetics of sex determination varies across taxa, sometimes even within a species. Major domesticated strains zebrafish ( Danio rerio ), including AB and TU, lack strong genetic determining locus, but more recently derived from nature, like Nadia (NA), possess ZZ male/ZW female chromosomal sex-determination system. fish pass through juvenile ovary stage, forming oocytes that survive in become females die males. To understand mechanisms gonad development NA zebrafish, we studied histology single cell transcriptomics developing ZW fish. developed by 22 days post-fertilization (dpf) directly formed testes, avoiding phase. Abstract The maternal-effect mutation ichabod ( ich ) results in ventralized zebrafish embryos due to impaired induction of the dorsal canonical Wnt-signaling pathway. While previous studies linked phenotype reduced ctnnb2 transcript levels, causative remained unidentified. Using long-read sequencing, we discovered that stems from insertion a non-autonomous CMC-Enhancer/Suppressor-mutator (CMC-EnSpm) transposon 3’UTR gene. Through reporter assays, demonstrate while wild type mRNAs exhibit remarkably high stability throughout early stages development, dramatically reduces stability. Genome-wide mapping CMC-EnSpm transposons across multiple strains also indicated ongoing transposition activity genome. Our findings not only resolve molecular basis but highlight continuing mutagenic potential endogenous and reveal unexpected aspects maternal regulation during development.

Small nucleolar RNAs (snoRNAs) are one of the most abundant and evolutionary ancient group functional non-coding RNAs. They were originally described as guides post-transcriptional rRNA modifications, but emerging evidence suggests that snoRNAs fulfil an impressive variety cellular functions. To reveal true complexity snoRNA-dependent functions, we need to catalogue first complete repertoire in a given context. While systematic mapping characterization "snoRNAomes" for some species have been recently, this has not done hitherto zebrafish (Danio rerio). Using size-fractionated RNA sequencing data from adult tissues, created interactive "snoRNAome" database species. Abstract Since the description of zebrafish Danio rerio in 1822, identity its closest living relative has been unclear. To address this problem, we sequenced exomes ten species genus and used them to infer relationships across 25 chromosomes genome. The majority within were remarkably consistent all chromosomes.
