Background: Plastic pollution driven by human activities has become a critical global issue for health. A growing literature demonstrates that micro- and nanoplastics (MNPs) contain endocrine-disrupting chemicals (EDCs) other harmful compounds enter the body easily, acting as agonists or antagonists wide range of hormonal receptors, promoting endocrine toxicity. Endocrine disruption induced MNPs occurs through aberrant activation/inhibition different signaling pathways in addition to directly interfering with balances, trigger apoptosis, oxidative stress, inflammation cells. However, date, molecular mechanisms these contaminants remain not completely elucidated. Furthermore, given unanimous consensus on negative impact health, several methodologies have been developed detect only environment but also biological fluids tissues. Results: This review comprehensively summarizes emerging experimental clinical evidence explaining underlying toxicity related chronic plastic relation system. In addition, illustrates new methodological approaches samples, highlighting employing complementary methods enables precise characterization quantification MNPs. Conclusions: Future studies experimental, epidemiological, epigenetic, multi-omics are essential understanding short long-term effects glands developing effective strategies mitigate their

Testicular aging is associated with declining reproductive health, but the molecular mechanisms are unclear. Here we generate a dataset of 214,369 single-cell transcriptomes from testicular cells 35 individuals aged 21–69, offering resource for studying and physiology. Machine learning analysis reveals stronger response in somatic compared to germ cells. Two waves aging-related changes identified: first peritubular donors their 30s, marked by increased basement membrane thickness, indicating priming state aging. In 50s, exhibit functional changes, including altered steroid metabolism Leydig immune responses macrophages. Further analyses reveal impact body mass index on spermatogenic capacity as age progresses, particularly after 45. Altogether, our findings illuminate alterations during testis relationship index, providing foundation future research potential diagnostic markers therapeutic targets. To enhance understanding human testis, authors provide map characteristics throughout lifespan. Using samples donors, they identify two examine interactions between index.

Abstract Background Infertility, which is defined as the inability to conceive after at least 12 months of regular unprotected sexual intercourses, affects about 15–20% couples worldwide and a male factor involved in half cases. The development assisted reproductive technology (ART) made it possible also individuals affected from severe oligospermia or azoospermia. However, impact on embryo development, implantation, prevalence chromosomal abnormalities, genetic epigenetic alterations, clinical obstetric outcomes still controversial. Purpose This narrative review examines indications, minimum access criteria, by individual ART technique relation factor.

The deleterious effects of chemical or non-chemical endocrine disruptors (EDs) on male fertility potential is well documented but still not fully elucidated. For example, the detection industrial chemicals' metabolites in seminal plasma and follicular fluid can affect efficiency gametogenesis, maturation competency gametes has guided scientists to hypothesize that disrupting chemicals (EDCs) may disrupt hormonal homoeostasis by leading a wide range control impairments. EDCs exposure reproductive health are highly dependent factors including type EDCs, duration exposure, individual susceptibility, presence other co-factors. In modern post-transition societies, we are reproducing later and living longer. While the impact of age on female reproductive function has been well studied, much less is known about intersection male reproduction. Our current understanding that advancing brings forth a progressive decline in fertility accompanied by reduction circulating testosterone levels appearance age-dependent pathologies including benign prostatic hypertrophy erectile dysfunction. Paternal ageing also associated with profound increase sperm DNA damage, multiple epigenetic changes germ line an elevated mutational load offspring. As global demographics shift toward increasing paternal age, the realm of assisted reproductive technologies (ARTs), particularly in vitro fertilization (IVF) and intracytoplasmic sperm injection (ICSI), faces new challenges opportunities. This study provides a comprehensive exploration implications advanced age on ART outcomes. Background research highlights social, cultural, economic factors driving men later fatherhood, with focus impact delayed paternity Methods involve thorough review existing literature, centering changes testicular function, semen quality, genetic epigenetic shifts associated advancing age. Study results point to intricate associations between father’s outcomes, older being linked diminished potential risks, varied impacts embryo implantation rates, birth The conclusions drawn from current suggest that while presents certain risks challenges, understanding mitigating these through strategies such as cryopreservation, lifestyle modifications, preimplantation testing can optimize Future directions are identified further comprehend mechanisms long-term effects father offspring health. underscores need for approach navigating intricacies fatherhood within context ART, aiming best possible outcomes couples their children.

Male fertility can be affected by oxidative stress (OS), which occurs when an imbalance between the production of reactive oxygen species (ROS) and body's ability to neutralize them arises. OS damage cells influence sperm production. High levels lipid peroxidation have been linked reduced motility decreased fertilization ability. This literature review discusses most commonly used biomarkers measure caused ROS, such as high level in seminal plasma indicator antioxidant activity. The investigated include 8-hydroxy-2-deoxyguanosine acid (8-OHdG), a marker DNA F2 isoprostanoids (8-isoprostanes) produced peroxidation. Furthermore, this focuses on recent methodologies including NGS polymorphisms differentially expressed gene (DEG) analysis, well epigenetic mechanisms ROS during spermatogenesis along with new developed evaluate biomarkers. Finally, addresses valuable insight into male infertility provided these advances how they led treatment possibilities. Overall, use has supplied innovative diagnostic therapeutic approaches, enhancing our understanding mechanisms.

Paternal programming is the concept that environmental signals from sire's experiences leading up to mating can alter semen and ultimately affect phenotype of resulting offspring. Potential mechanisms carrying paternal effects offspring be associated with epigenetic signatures (DNA methylation, histone modification non-coding RNAs), oxidative stress, cytokines, seminal microbiome. Several opportunities exist for sperm/semen influenced during development; these are within testicle, epididymis, or accessory sex glands. Epigenetic sperm impacted pre-natal pre-pubertal periods, sexual maturity advancing sire age.
