SUMMARY Staphylococcus aureus is an important cause of skin and soft-tissue infections (SSTIs), endovascular infections, pneumonia, septic arthritis, endocarditis, osteomyelitis, foreign-body sepsis. Methicillin-resistant S. (MRSA) isolates were once confined largely to hospitals, other health care environments, patients frequenting these facilities. Since the mid-1990s, however, there has been explosion in number MRSA reported populations lacking risk factors for exposure system. In recent years, methicillin-resistant Staphylococcus aureus (MRSA) have become a truly global challenge. addition to the long-known healthcare-associated clones, novel strains also emerged outside of hospital settings, in community as well livestock. The emergence and spread virulent clones expressing Panton-Valentine leukocidin (PVL) is an additional cause for concern. order provide overview pandemic, epidemic sporadic strains, more than 3,000 clinical veterinary isolates MRSA mainly from Germany, United Kingdom, Ireland, France, Malta, Abu Dhabi, Hong Kong, Australia, Trinidad & Tobago some reference States been genotyped by DNA microarray analysis. This technique allowed assignment 34 distinct lineages which can be clearly defined based on non-mobile genes. results were accordance with data multilocus sequence typing. More 100 different distinguished affiliation these lineages, SCCmec type presence or absence PVL. Methicillin resistance in staphylococci is mediated by penicillin binding protein 2a (PBP 2a), encoded mecA on mobile staphylococcal cassette chromosome mec (SCCmec) elements. In this study, two clonal complex 130 (CC130) methicillin-resistant Staphylococcus aureus (MRSA) isolates from patients Irish hospitals were identified that phenotypically PBP positive but lacked conventional PCR and DNA microarray screening. The as methicillin-susceptible S. using the GeneXpert real-time assay. Whole-genome sequencing of one isolate (M10/0061) revealed a 30-kb SCCmec element encoding class E with highly divergent blaZ-mecA-mecR1-mecI, type 8 recombinase (ccr) consisting ccrA1-ccrB3, an arsenic operon, flanking direct repeats (DRs). was almost identical to XI (SCCmec XI) Sanger Institute sequence 425 bovine MRSA strain LGA251 listed website International Working Group Classification Staphylococcal Cassette Chromosome Elements. open reading frames (ORFs) within M10/0061 exhibited 21 93% amino acid identity ORFs GenBank.
