Melatonin, a pleiotropic neurohormone, is recognized for its hepatoprotective role in metabolic dysfunction-associated steatotic liver disease (MASLD) triggered by diet and/or chronodisruption. Nocturnin (Noct), circadian clock output regulating hepatic lipid metabolism, has an unclear MASLD. This study explores oscillations of Noct MASLD and synergy with melatonin. Differential gene expression analysis identified key lipogenic genes (HNF-4A SCD) as targets enzymatic activity. Male C57BL/6J mice were subjected to high-fat-high-fructose (H) or without chronodisruption (CD) 16 weeks. mRNA peaked at ZT12 Control H groups, while CD HCD groups showed peaks ZT6, alongside increased % relative amplitude, cumulative expression, shifted Centre Gravity (CoG). Melatonin treatment H, CD, significantly reduced amplitude expression. In OA-treated HepG2 cells, protein levels rose 36 h 32 h, respectively, CoG shifts. but correcting rhythmicity. Collectively, induces elevation phase shifts melatonin demonstrates corrective effects on phasing, underscoring therapeutic potential. .

Abstract Thyroid hormones (THs) are important regulators of systemic energy metabolism. In the liver, they stimulate lipid and cholesterol turnover increase bioavailability. It is still unknown how TH state interacts with circadian clock, another regulator We addressed this question using a mouse model hypothyroidism performed analyses. Low levels decreased locomotor activity, food intake, body temperature mostly in active phase. Concurrently, liver transcriptome profiling showed only subtle effects compared to elevated conditions. Comparative revealed alterations mesor, amplitude, phase transcript livers low-TH mice. Genes associated uptake, biosynthesis, bile acid secretion reduced mesor. Increased serum were identified, respectively. Combining data from low- high-TH conditions allowed identification 516 genes mesor changes as molecular markers state. explored these created an expression panel that assesses time-of-day dependent manner. Our findings suggest has low action under physiological Circadian reveals potential

The circadian system is an intricate molecular network of coordinating clocks that organize the internal synchrony organism in response to environment. These rhythms are maintained by genetically programmed positive and negative auto-regulated transcriptional translational feedback loops sustain 24-hour oscillations mRNA protein components endogenous clock. Since inter intracellular activity central pacemaker appears reduce with aging, interaction between clock aging continues elude our understanding. In this review article, we discuss at level how adversely affects functioning rodents humans. natural decline melatonin levels strongly contributes dysregulation resulting development neurological anomalies. Additionally, inappropriate environmental conditions such as Artificial Light Night (ALAN) can cause disruption or chronodisruption (CD) which result a variety pathological diseases, including premature aging. Furthermore, summarize recent evidence suggesting CD may also be predisposing factor for age-related neurodegenerative diseases (NDDs) Alzheimer's disease (AD), Parkinson's (PD), Huntington's (HD), although more investigation required prove link. Finally, certain chrono-enhancement approaches have been offered intervention strategies prevent, alleviate, mitigate impacts CD. Many animals benefit from synchronizing their daily activities with conspecifics. In this hybrid paper, we first review recent literature supporting and extending earlier evidence for a lack of clear relationship between the level sociality social entrainment circadian rhythms. Social is specifically potent in that live constant environments which some or all individuals do not experience ambient day-night cycles. We next focus on highly honeybees there good cues entrain clocks nest bees can override influence conflicting light-dark The current understanding synchronization consistent self-organization models surrogates forager activity, such as substrate-borne vibrations colony volatiles, dwelling dark cavity nest. Rationale: Mechanical ventilation (MV) is life-saving but may evoke ventilator-induced lung injury (VILI). Objectives: To explore how the circadian clock modulates severity of murine VILI via core component BMAL1 (basic helix-loop-helix ARNT like 1) in myeloid cells. Methods: Myeloid cell BMAL1-deficient (LysM (lysozyme 2 promoter/enhancer driving cre recombinase expression)Bmal1-/-) or wild-type control (LysMBmal1+/+) mice were subjected to 4 hours MV (34 ml/kg body weight) induce injury. Ventilation was initiated at dawn dusk complete darkness (circadian time [CT] 0 CT12) determine diurnal and effects. Lung quantified by function, pulmonary permeability, blood gas analysis, neutrophil recruitment, inflammatory markers, histology. Neutrophil activation oxidative burst analyzed ex vivo. Measurements Main Results: In experiments, ventilated exhibited higher permeability recruitment compared with dusk. Experiments CT showed deterioration worsening oxygenation, increased mortality CT0 CT12. Wild-type neutrophils isolated reactive oxygen species production dusk, whereas these day-night differences dampened LysMBmal1-/- neutrophils. mice, variations VILI-induced reduced LysMBmal1+/+ mice. Conclusions: Inflammatory response barrier dysfunction upon exhibit variations, regulated clock. are less susceptible ventilation-induced pathology lack variation Our data suggest that internal cells an important modulator VILI.

Abstract Background Glucagon is secreted from pancreatic alpha cells in response to low blood glucose and increases hepatic production. Furthermore, glucagon enhances protein lipid metabolism during a mixed meal. Glucagon-like peptide-1 (GLP-1) glucose-dependent insulinotropic polypeptide (GIP) are gut endocrine meals control homeostasis by potentiating insulin secretion inhibiting food intake. Both intake have been reported be affected circadian rhythms vice versa. In this study, we investigated whether the of glucagon, GLP-1 GIP was rhythms. Methods A total 24 healthy men with regular sleep schedules were examined for h at hospital ward 15 wakefulness 9 sleep. Food standardized, samples obtained every third hour. Plasma concentrations measured, data analyzed rhythmometric statistical methods. Available on plasma C-peptide also included. Results GLP-1, GIP, fluctuated diurnal 24-h rhythm, highest levels day lowest night: ( p < 0.0001, peak time 18:26 h), 17:28 18:01 17.59 23:26 h). As expected, found significant correlations between but did not find GIP. Conclusions Our results demonstrate that under meal conditions similar many free-living individuals, observed higher daytime evening than overnight. These findings underpin disturbed rhythm as potential risk factor diabetes obesity. Trial registration ClinicalTrials.gov Identifier: NCT06166368. Registered 12 December 2023.

In this study, we have investigated role of circadian rhythm signaling pathway, a key regulator human health, in pathogenesis HTLV-1 using Boolean Network analysis laboratory methods. After extensive search analyzed relationships between genes pathway R programming language BoolNet package. Subsequently, examined impact viral proteins on clock genes. Finally, identified three genes, PER2, CRY1, DEC1, as main checkpoints from attractors obtained. These two were quantitatively assessed groups individuals, ten asymptomatic carriers infected with healthy individuals qRT-PCR method.
