Macrophages
are
crucial
members
of
the
mononuclear
phagocyte
system
essential
to
protect
host
from
invading
pathogens
and
central
inflammatory
response
with
their
ability
acquire
specialized
phenotypes
(M1)
anti-inflammatory
(M2)
produce
a
pool
mediators.
Equipped
broad
range
receptors,
such
as
Toll-like
receptor
4
(TLR4),
CD14,
Fc
gamma
receptors
(Fc
γ
Rs),
macrophages
can
efficiently
recognize
phagocytize
secrete
cytokines
by
triggering
various
secondary
signaling
pathways.
Phospholipase
C
(PLC)
is
family
enzymes
that
hydrolyze
phospholipids,
most
significant
which
phosphatidylinositol
4,5-bisphosphate
[PI(4,5)P2].
Cleavage
at
internal
phosphate
ester
generates
two
second
messengers,
inositol
1,4,5-trisphosphate
(IP3)
diacylglycerol
(DAG),
both
mediate

cellular
functions
including
response.

Lactobacilli
have

shown
to
promote
health
functions.
In
this
study,
we
analyzed

mechanism

probiotics
affected

immunity,

represent

approach

Monocytes
and
macrophages
are
essential
components
of
innate
immune
system
have
versatile
roles
in
homeostasis
immunity.
These
phenotypically
distinguishable
mononuclear
phagocytes
play
distinct
different
stages,
contributing
to
the
pathophysiology

them

potentially

therapeutic

conditions.
Several

evidence
supported

receptors.
Macrophage-colony
stimulating
factor
(CSF-1)
interleukine-34
(IL-34),
also
known
as
"twin"
cytokines,
part
this
regluatory
landscape.
CSF-1
IL-34
share
common
receptor,
macrophage-colony
receptor
(CSF-1R),
is
activated
in
similar
way
both

signaling
pathways.
However,
there
some
discrete
differential
activation

to
activities.
In
review,
we
disscuss
recent
progress
understanding
role
twin
cytokines
differentiation,
from
interaction
CSF-1R
pathways,
implication
macrophages.
A
special
focus
IL-34,
its
involvement
pathophsyiological
contexts,
potential
theranostic
target
for
therapy
will
be
proposed.
