Macrophages are crucial members of the mononuclear phagocyte system essential to protect host from invading pathogens and central inflammatory response with their ability acquire specialized phenotypes (M1) anti-inflammatory (M2) produce a pool mediators. Equipped broad range receptors, such as Toll-like receptor 4 (TLR4), CD14, Fc gamma receptors (Fc γ Rs), macrophages can efficiently recognize phagocytize secrete cytokines by triggering various secondary signaling pathways. Phospholipase C (PLC) is family enzymes that hydrolyze phospholipids, most significant which phosphatidylinositol 4,5-bisphosphate [PI(4,5)P2]. Cleavage at internal phosphate ester generates two second messengers, inositol 1,4,5-trisphosphate (IP3) diacylglycerol (DAG), both mediate in diverse cellular functions including response. Recent studies have shown some PLC isoforms involved multiple stages TLR4-, CD14-, Rs-mediated activation nuclear factor kappa B (NF- κ B), mitogen-activated protein kinase (MAPK), interferon regulatory factors (IRFs), all associated regulation Therefore, implicated pathogenesis numerous diseases. This review provides an overview our current knowledge on how regulates macrophage-mediated

Lactobacilli have been shown to promote health functions. In this study, we analyzed the mechanism by which four different strains of probiotics affected innate immunity, such as regulation ROS, cytokines, phagocytosis, bactericidal activity, signaling NF-κB pp65, and TLR2 activation. The production ROS was dependent on concentration species Lactobacillus. results obtained from tested (Lactobacillus rhamnosus GG, L. KLSD, helveticus IMAU70129, casei IMAU60214) showed that induced early proinflammatory cytokines IL-8,TNF-α, IL-12p70, IL-6. However, IL-1β expression only (after 24 h stimulation). Phagocytosis activity macrophages against various pathogens, S. aureus, typhimurium, E. coli, were increased pretreatment with nuclear translocation pp65 TLR2-dependent also treatment probiotics. Taken together, experiments demonstrate probiotic Lactobacillus exert immunostimulatory effects may be directly linked initial inflammation response human macrophages.

Cellular immunotherapies represent a promising approach for the treatment of cancer. Engineered adoptive cell therapies redirect and augment leukocyte’s inherent ability to mount an immune response by introducing novel anti-tumor capabilities targeting moieties. A prominent example this is use T cells engineered express chimeric antigen receptors (CARs), which have demonstrated significant efficacy against some hematologic malignancies. Despite increasingly sophisticated strategies harness function, solid tumors has remained elusive therapies. Amongst types used in immunotherapies, however, macrophages recently emerged as candidates tumors. In review, we discuss monocytes Macrophages are innate that intrinsically equipped with broad therapeutic effector functions, including active trafficking tumor sites, direct phagocytosis, activation microenvironment professional presentation. We focus on engineering manipulating macrophages, specific CAR (CAR-M). highlight design production CAR-M transfer, clinical considerations their treating then outline recent progress results applying immunotherapies. The development macrophage-based holds promise key weapon therapy armamentarium.

Monocytes and macrophages are essential components of innate immune system have versatile roles in homeostasis immunity. These phenotypically distinguishable mononuclear phagocytes play distinct different stages, contributing to the pathophysiology various forms making them a potentially attractive therapeutic target inflammatory conditions. Several pieces evidence supported role cell surface receptors expressed on these cells their downstream signaling molecules initiating perpetuating response. In this review, we discuss current understanding monocyte macrophage biology inflammation, highlighting chemoattractants, inflammasomes, integrins function monocytes during events inflammation. This review also covers recent interventions targeting at cellular molecular levels.

Macrophages are specialized cells that control tissue homeostasis. They include non-resident and tissue-resident macrophage populations which characterized by the expression of particular cell surface markers secretion molecules with a wide range biological functions. The differentiation polarization macrophages relies on specific growth factors their receptors. Macrophage-colony stimulating factor (CSF-1) interleukine-34 (IL-34), also known as "twin" cytokines, part this regluatory landscape. CSF-1 IL-34 share common receptor, macrophage-colony receptor (CSF-1R), is activated in similar way both turns identical signaling pathways. However, there some discrete differential activation leading to activities. In review, we disscuss recent progress understanding role twin cytokines differentiation, from interaction CSF-1R pathways, implication macrophages. A special focus IL-34, its involvement pathophsyiological contexts, potential theranostic target for therapy will be proposed.
