Ginseng, when used as a food and nutritional supplement, has the ability to regulate human immunity. Here, potential anti-hepatic fibrosis effect of ginsenoside Rd (Rd), one protopanaxadiol types ginsenoside, was investigated. We established hepatic model using intraperitoneal injection thioacetamide (TAA) for five weeks in mice. In addition, an vitro by TGF-β activate stellate cells (HSCs), treated with estrogen-related receptor α (ERRα) inhibitor (XCT-790). The ERRα knockdown (shRNA-ERRα) primary mouse hepatocytes establish hepatocyte injury TGF-β, they were then incubated Rd. significantly alleviated histopathological changes, reduced serum alanine aminotransferase (ALT) aspartate (AST) levels. could upregulate downregulate markers livers TAA-induced mice, inhibited purinergic ligand-gated ion channel 7 (P2X7r)-mediated NLRP3 inflammasome activation, consequently reversing liver inflammatory response. increased expression suppressed extracellular matrix (ECM) HSCs or hepatocytes. decreased P2X7r-mediated response, including production IL-1β, IL-23 activated ameliorate damage further inhibit entry IL-1β IL-18 into matrix. reaction regulating ERRα-P2X7r signaling pathway while suppressing fibrogenesis, which suggests that can serve novel dietary supplement approach combat fibrosis.

Depression is a mental health disorder that develops as result of complex psycho-neuro-immuno-endocrinological disturbances. This disease presents with mood disturbances, persistent sadness, loss interest and impaired cognition, which causes distress to the patient significantly affects ability function have satisfying family, social professional life. requires comprehensive management, including pharmacological treatment. Because pharmacotherapy depression long-term process associated risk numerous adverse drug effects, much attention paid alternative therapy methods, phytopharmacotherapy, especially in treating mild or moderate depression. Preclinical studies previous clinical confirm antidepressant activity active compounds plants, such St. John’s wort, saffron crocus, lemon balm lavender, less known European ethnopharmacology, roseroot, ginkgo, Korean ginseng, borage, brahmi, mimosa tree magnolia bark. The these plants exert antidepressive effects similar mechanisms those found synthetic antidepressants. description phytopharmacodynamics includes inhibiting monoamine reuptake oxidase complex, agonistic antagonistic on multiple central nervous system (CNS) receptors. Moreover, it noteworthy anti-inflammatory effect also important mentioned above light hypothesis immunological disorders CNS are significant pathogenetic factor narrative review results from traditional, non-systematic literature review. It briefly discusses pathophysiology, symptomatology treatment depression, particular focus role phytopharmacology its provides action revealed experimental ingredients isolated herbal antidepressants selected confirming their effectiveness.

Depressive disorder is a common mental characterized by depressed mood and loss of interest or pleasure. As the Herbal medicines are mainly used as complementary alternative therapy for depression. This study aimed at exploring antidepressant activity Huang-lian Jie-du Decoction (HLJDD), evaluating active components potential depression-associated targets. HLJDD was administered on chronic unpredictable mild stress-induced (CUMS) depressive mice. Behavior evaluation performed through force swimming test (FST), novelty-suppressed feeding (NSF), open field (OFT). Active HLJDD, targets, metabolic pathways involved in depression were explored systemic biology-based network pharmacology assay, molecular docking metabonomics. FST assay showed that CUMS mice with had significantly shorter immobility time compared control Further, alleviated latency NSFand increased moving distance duration OFT. In following thirty-eight compounds identified based drug-like characteristics, pharmacokinetics pharmacodynamics profiles. Moreover, forty-eight targets ten biochemical uncovered GRIN2B, DRD, PRKCA, HTR, MAOA, SLC6A4, GRIN2A, CACNA1A implicated inhibition symptoms modulating tryptophan metabolism, serotonergic dopaminergic synaptic activities, cAMP signaling pathway, calcium pathway. Further pharmacology-based analysis correlation between metabolism. A total thirty-seven compounds, seventy-six sixteen These findings show acts such INS, MAO, CAT, FoxO, PI3K/Akt, calcium, HIF-1, mTOR pathways, modulates serotoninergic functions. addition, metabonomics metabolism primary target The exhibited effects. SLC6A4 MAOA modulated berberine, baicalein, tetrahydroberberine, candicine may be main HLJDD.

Medicinal plants and their extracts are natural remedies with enormous potential for treating various diseases, including depression anxiety. In the case of depression, hundreds have traditionally been used in folk medicine generations. Different plant products analyzed as antidepressant agents validated models to test antidepressant-like effects animals, although other complementary studies also employed. Most these focus on possible mediators implicated effects, dopamine, serotonin, noradrenaline being principal neurotransmitters implicated, both through interference receptors metabolism by monoamino oxidases, well neuro-endocrine neuroprotective effects. There approximately 650 reports medicinal PubMed; 155 them compiled this review, a relevant group yielding positive results. Saffron turmeric most species studied preclinical clinical studies; St. John's wort or kava tested extensively. L-tryptophan (Trp) contributes to regulating bilateral communication of the gut-brain axis. It undergoes three major metabolic pathways, which lead formation kynurenine, serotonin (5-HT), and indole derivatives (under control microbiota). Metabolites from principal Trp pathway, kynurenic acid quinolinic acid, exhibit neuroprotective activity, while picolinic exhibits antioxidant 5-HT modulates appetite, sleep cycle, pain. Abnormality in plays crucial roles diseases, including depression, colitis, ulcer, gut microbiota-related dysfunctions. To address these use natural products could be a favorable alternative because they are rich source compounds that can modulate activity combat various diseases through modulating different signaling microbiota, kynurenine pathway. Alterations cascade pathways via phytochemicals may help us explore deep relationships axis study neuroprotection. This review highlights their metabolites targeting diseases. Additionally, role regulation gastroprotective activities is discussed. compiles literature on novel, potent agents action mechanisms proposes prospective future studies identify more pharmaceuticals based Trp.
