Atopic dermatitis (AD) is a globally prevalent chronic inflammatory skin disorder. Its pathogenesis remains incompletely understood, resulting in considerable therapeutic challenges. Recent studies have highlighted the significance of interaction between AD and gut microbiome. In this study, we investigated effects probiotic-derived extracellular vesicles on AD. Initially, isolated characterized from Limosilactobacillus fermentum SLAM 216 (LF216EV) their composition through multi-omics analysis. Gene ontology (GO) pathway analysis classified LF216EV proteins into biological processes, molecular functions, cellular components. Importantly, specific abundance linoleic, oleic, palmitic, sebacic, stearic acids indicating upregulated fatty acid metabolism were observed by metabolomic Furthermore, featured lipid profiling including AcylGlcADG ceramide LF216EV. an atopic dermatitis-like cell model induced TNFα/IFNγ, significantly modulated expression immune regulatory genes (TSLP, TNFα, IL-6, IL-1β, MDC), its potential functionality dermatitis. alleviated AD-like phenotypes, such as redness, scaling/dryness, excoriation, DNCB. Histopathological revealed that decreased epidermal thickness mast infiltration dermis. administration reduced mouse scratching depression-related behaviors, with faster onset than classical treatment dexamethasone. quantitative real-time polymerase chain reaction (qRT-PCR) analysis, significant increase levels htrb2c, sert, tph-1, associated serotonin, LF216EV-treated group, along total serum serotonin levels. Gut microbiome group altered microbiota profile. Correlation genera Desulfovibrio differences intestinal metabolites, serotonin. Our findings demonstrate mitigates symptoms promoting synthesis modulation metabolome composition.

The chronic stress of social isolation is a valid predictor cognitive pathology. This study aimed to compare the effects long-term on female versus male Wistar rats' learning and memory. We hypothesized that prolonged stress, which starts early in life, would affect sex-dependent manner.Social started at edge mid-adolescence lasted 9 months. rat's abilities were assessed by habituation reactivity novelty open field (OF) test, spatial memory Morris water maze (MWM), conditioned passive avoidance (PA) reflex. Basal serum corticosterone levels using an enzyme-linked immunosorbent assay.Regardless housing conditions, females habituated OF under low illumination slower than males. Under bright light, single-housed rats showed hyporeactivity novelty. In MWM, all learned locate platform; however, first training day, females' speed was lower relative other groups. Four months later, post-reminder probe trial, reached area around platform site only males, regardless preferred target quadrant. Single-housed rats, irrespective sex, PA deficit. There more pronounced fear males females. both basal rat blood after did not differ from group-housed corresponding sex. Meanwhile, level higher conditions. weight adrenal glands increased females.Under isolation, compared with impairments MWM paradigm, findings specify their greater vulnerability isolation.

Abstract Background Experimental mice are often single-housed either for an individual analysis (feeding behavior, imaging, calorimetry) or as a stress paradigm (social isolation) in translational biomedical research. Reports of the influence single housing rodents conflicting and may depend on age duration isolation. Sex is not included factor. In this study we investigated effects 4-week male female body weight, serum corticosterone levels. Results Behavioral tests showed no effect anhedonia coping, anxiety motor exploration. Social avoidance occurred both males females. Regarding physiological effects, did induce changes levels, but reduced weight gain. Conclusions While some mouse studies chronic social isolation reported depression-related disturbances, our data suggest that might be necessarily too stressful. This important animal welfare regulations experiments life science

Alzheimer's disease (AD) is the leading cause of cognitive impairment and dementia in elderly individuals. According to current biomarker framework for “unbiased descriptive classification”, biomarkers neurodegeneration, “N”, constitute a critical component tri‐category “A/T/N” system. Current neurodegeneration suffer from potential drawbacks such as requiring invasive lumbar puncture, involving ionizing radiation, or representing late, irreversible marker. Recent human studies have suggested that reduced brain oxygen metabolism may be new functional marker AD, but heterogeneity presence mixed pathology patients did not allow full understanding role extraction AD. In this report, global related physiological parameters were studied two AD mouse models with relatively pure pathology, using advanced MRI techniques including T 2 ‐relaxation‐under‐spin‐tagging (TRUST) phase contrast (PC) MRI. Additionally, regional cerebral blood flow (CBF) was determined pseudocontinuous arterial spin labeling. Reduced fraction (by −18.7%, p = 0.008), unit‐mass metabolic rate (CMRO ) −17.4%, 0.04) total CMRO −30.8%, < 0.001) observed Tau4RΔK mice—referred tau model—which manifested pronounced measured by diminished volume −15.2%, 0.001). Global CBF these mice different those wild‐type ( > 0.05), suggesting normal vascular function. By contrast, B6;SJL‐Tg [APPSWE]2576Kha (APP) amyloid model—no reduction, well intact metabolism, found 0.05). Consistent imaging data, behavioral measures walking distance impaired 0.004), APP 0.88). For group-living animals, the social environment provides salient experience that can weaken or strengthen aspects of cognition such as memory recall. Although cellular substrates individually acquired fear memories in dentate gyrus (DG) and basolateral amygdala (BLA) have been well-studied recent work has revealed circuit mechanisms underlying encoding experience, processes by which interacts with an individual’s to alter recall remain unknown. Here we show stressful experiences enhance previously male but not female mice, buffering conspecifics’ distress blocks this enhancement. Activity-dependent tagging cells DG during learning these ensembles were endogenously reactivated males, even after extinction. These shown be functional components engrams, optogenetic stimulation active fear-conditioned naïve animals was sufficient drive fear-related behaviors. Taken together, our findings suggest reactivate preexisting engrams thereby discrete memories.

Animal welfare is recognized as essential for the coexistence of humans and animals. Considering increased demand interest in animal welfare, many methods improving are being devised, but which method reduces stress has not been scientifically verified. Therefore, reducing by providing a proper breeding environment environmental enrichment can be basis study. In this study, levels were assessed based on mouse-breeding environment. We considered that higher body weight lower corticosterone concentration, stress. According to results, animals individual ventilation cages determined have serum cortisol concentrations, while was when compared with isolated group or enrichment. The results provide appropriate guidelines laboratory welfare.
