The intestinal epithelial barrier (IEB) is one of the largest interfaces between environment and internal milieu body. It essential to limit passage harmful antigens microorganisms and, on other side, assure absorption nutrients water. maintenance this delicate equilibrium tightly regulated as it for human homeostasis. Luminal solutes ions can pass across IEB via two main routes: transcellular pathway or paracellular pathway. Tight junctions (TJs) are a multi-protein complex responsible regulation permeability. TJs control through have key role in maintaining integrity. Several factors, including cytokines, gut microbiota, dietary components known regulate TJs. Gut microbiota participates several functions modulation cells immune system release metabolites, such short-chain fatty acids (SCFAs). Mediators released by induce cell damage dysfunction. subsequent disruption allows into mucosa leading further inflammation. Growing evidence indicates that dysbiosis, activation, dysfunction diseases, irritable bowel syndrome (IBS), inflammatory disease (IBD), gluten-related conditions. Here we summarize interplay mucosal their involvement IBS, IBD, disorders.

Interleukin (IL)-18 was originally discovered as a factor that enhanced IFN-γ production from anti-CD3-stimulated Th1 cells, especially in the presence of IL-12. Upon stimulation with Ag plus IL-12, naïve T cells develop into IL-18 receptor (IL-18R) expressing which increase response to stimulation. Therefore, IL-12 is commitment induces development cells. In contrast, proinflammatory cytokine facilitates type 1 responses. However, without but IL-2, stimulates NK CD4+ NKT and established produce IL-3, IL-9, IL-13. Furthermore, together mast basophils IL-4, IL-13, chemical mediators such histamine. various cell types has pleiotropic functions. member IL-1 family cytokines. demonstrates unique function by binding specific expressed on this review article, we will focus features health disease experimental animals humans.

Mucus is a selective barrier to particles and molecules, preventing penetration the epithelial surface of mucosal tissues. Significant advances in transmucosal drug delivery have recently been made emphasized that an understanding basic structure, viscoelastic properties, interactions mucus great value design efficient systems. Mucins, primary non-aqueous component mucus, are polymers carrying complex heterogeneous structure with domains undergo variety molecular interactions, such as hydrophilic/hydrophobic, hydrogen bonds electrostatic interactions. These properties directly relevant numerous mucin-associated diseases, well delivering drugs across barrier. Therefore, this review we discuss regional differences composition, physicochemical pore size, viscoelasticity, pH, ionic strength. factors also discussed respect changes function disease state. Collectively, seeks provide state art roadmap for researchers who must contend critical delivery.

In this contribution, we review the molecular and physiological barriers to oral delivery of peptides nanoparticles. We discuss opportunities predictivity various in vitro systems with special emphasis on human intestine Ussing chambers. First, constraints peptide absorption are discussed. Then examined. These include gastric intestinal environment, mucus barrier, tight junctions between epithelial cells, enterocytes epithelium, subepithelial tissue. Recent data from proteome studies used provide information about protein expression profiles different nanoparticle absorption. Strategies that have been employed increase across each Special consideration is given attempts at utilizing endogenous transcytotic pathways. To reliably translate or permeability vivo situation a subject, experimental system needs realistically capture central aspects mentioned barriers. Therefore, characteristics common cell culture discussed compared those tissues. Attempts use tissue models for vitro–in extrapolation reviewed.

Abstract The intestine is the largest interface between internal body and external environment. intestinal barrier a dynamic system influenced by composition of microbiome activity intercellular connections, regulated hormones, dietary components, inflammatory mediators, enteric nervous (ENS). Over years, it has become increasingly evident that maintaining stable crucial to prevent various potentially harmful substances pathogens from entering Disruption referred as 'leaky gut' or leaky gut wall syndrome seems be characterized release bacterial metabolites endotoxins, such lipopolysaccharide (LPS), into circulation. This condition, mainly caused infections, oxidative stress, high-fat diet, exposure alcohol chronic allergens, dysbiosis, appear highly connected with development and/or progression several metabolic autoimmune systemic diseases, including obesity, non-alcoholic fatty liver disease (NAFLD), neurodegeneration, cardiovascular disease, bowel type 1 diabetes mellitus (T1D). In this review, starting description mechanisms enable homeostasis analyzing relationship complex ecosystem pathological conditions, we explore role in driving inflammation, also shedding light on current future therapeutic interventions.
