Abstract
Objective
Proliferative
verrucous
leukoplakia
(PVL)
has
high
rates
of
malignant
transformation
into
oral
squamous
cell
carcinoma
(OSCC),
but
the
clinical
and
evolutionary
pattern
OSCC
from
PVL
(PVL‐OSCC)
is
more
favorable
than
that
not
preceded
by
(OSCC).
Here,
we
aimed
to
explore
pathophysiologic
differences
between
PVL‐OSCC
through
transcriptomic
DNA
methylation
analyses.
Materials
methods
In
this
case‐control
study,
biopsies
8
10
patients
were
obtained
for
global
sequencing
using
RNAseq
a
genome‐wide
analysis
via
Infinium
EPIC
Platform
(graphical
abstract).
Results
One
hundred
thirty‐three
differentially
expressed
genes
(DEGs)
detected,
94
them
upregulated
in
OSCC.
Most
these
previously
described
cancer
associated
with
prognosis.
The
integrative
revealed
26
DEGs,
corresponding
37
CpGs,
whose
promoters
regulated
methylation.
Twenty‐nine
CpGs
found
as
hypermethylated
PVL‐OSCC.
Only
5
aberrantly
methylated
patients,
whereas
21
underexpressed.
Conclusions
presented
lower
expression
cancer‐related
genes.
Hypermethylation
promoter
region
many
was
also
noticed,
indicating
could
be
regulatory
mechanism.
Proliferative
verrucous
leukoplakia
(PVL),
distinguished
by
its
malignant
transformation
rate
of
43.87%
to
65.8%,
stands
as
the
oral
potentially
disorder
with
highest
propensity
for
malignancy.
PVL
is
marked
distinctive
heterogeneity
regarding
clinical
or
histopathological
characteristics
well
prognostic
factors
pertinent
this
condition.

Proliferative
verrucous
leukoplakia
(PVL)
is
considered
as
an
oral
potentially
malignant
disorder
(OPMD)
that
presents
with
a
high
tendency
to
recurrence
after
treatment
and
has
the
highest
transformation
ratio
among
all
OPMD
(50%).
Evidence-based
publications
have
indicated
evolution
reported
significantly
related
inconsistent
diagnostic
criteria
used
in
primary-level
studies;
so,
it
been
hypothesized
risk
of
cancer
for
this
disease
could
even
be
underestimated.

because
PVL
requires
specific
management
protocols,
evidence-based,

early
