The skin is the largest organ of body and consists an epidermis, dermis subcutaneous adipose tissue. surface area often stated to be about 1.8 2 m2 represents our interface with environment; however, when one considers that microorganisms live in hair follicles can enter sweat ducts, interacts this aspect environment becomes 25–30 m2. Although all layers skin, including tissue, participate antimicrobial defense, review will focus mainly on role factors epidermis at surface. outermost layer stratum corneum, physically tough chemically inert which protects against numerous environmental stresses. It provides a permeability barrier attributable lipids intercellular spaces between corneocytes. In addition barrier, there innate involves lipids, peptides proteins. has low pH poor certain nutrients, limits range survive there. Melanin trans-urocanic acid provide protection from UV radiation, Langerhans cells are poised monitor local trigger immune response as needed. Each these protective barriers discussed.

Abstract Skin wounds are common and represent a major public health economical problem, with risks of complications significant negative impact on the quality life patients. Cutaneous wound healing is tightly regulated process resulting in restoration tissue integrity. Wound involves interaction several skin, immune vascular cells, growth factors cytokines. However, external actors can play an important role healing, such as skin microbiome, which microbial commensal collection bacteria, fungi viruses inhabiting skin. Indeed, recent advances have featured interactions, within environment, between different species host system. This article reviews relationship microbiome process. Although cutaneous potential entry site for infection, either detrimental or beneficial healing. Thus, targeting could essential part management.

The goal of the current work was to create an antibacterial agent by using polycaprolactone/chitosan (PCL/CH) nanofibers loaded with Cordia myxa fruit extract (CMFE) as antimicrobial for wound dressing. Several characteristics, including morphological, physicomechanical, and mechanical surface wettability, activity, cell viability, in vitro drug release, were investigated. inclusion CMFE PCL/CH led increased swelling capability maximum weight loss. SEM images PCL/CH/CMFE mat showed a uniform topology free beads average fiber diameter 195.378 nm. Excellent activity shown towards Escherichia coli (31.34 ± 0.42 mm), Salmonella enterica (30.27 0.57 Staphylococcus aureus (21.31 0.17 Bacillus subtilis (27.53 1.53 Pseudomonas aeruginosa (22.17 0.12 mm) based on inhibition zone assay. sample containing 5 wt% had lower water contact angle (47 3.7°), high porosity, compared neat mat. release 5% CMFE-loaded proven be anomalous non-Fickian diffusion Korsmeyer-Peppas model. Compared pure PCL membrane, PCL-CH/CMFE membrane exhibited suitable cytocompatibility L929 cells. In conclusion, fabricated nanofibrous films demonstrated bioavailability, properties that can used dressings.

Lipid synthesis is necessary for formation of epithelial barriers and homeostasis with external microbes. An analysis the response human keratinocytes to several different commensal bacteria on skin revealed that Cutibacterium acnes induced a large increase in essential lipids including triglycerides, ceramides, cholesterol, free fatty acids. A similar occurred mouse epidermis affected acne. Further showed this was mediated by short-chain acids produced dependent increased expression lipid genes glycerol-3-phosphate-acyltransferase-3. Inhibition or RNA silencing peroxisome proliferator-activated receptor-α (PPARα), but not PPARβ PPARγ, blocked response. The keratinocyte content improved innate barrier functions antimicrobial activity, paracellular diffusion, transepidermal water loss. These results reveal metabolites from common bacterium have previously unappreciated influence composition epidermal lipids.

In recent years, significant progress has been made in transdermal drug delivery systems, but there is still a search for enhancers that can improve the absorption of active substances through stratum corneum. Although permeation have described scientific literature, use naturally occurring this role particular interest, because they offer high level safety use, with low risk skin irritation, and efficiency. addition, these ingredients are biodegradable, easily available, widely accepted by consumers due to growing trust natural compounds. This article provides information on derived compounds systems help them penetrate skin. The work focuses components found corneum such as sterols, ceramides, oleic acid, urea. Penetration nature, mainly plants, terpenes, polysaccharides, fatty acids also described. mechanism action discussed, methods assessing their penetration efficiency provided. Abstract The human body harbors a diverse ecosystem of microorganisms, including bacteria, viruses, and fungi, collectively known as the microbiota. Current research is increasingly focusing on potential association between microbiota various neuropsychiatric disorders. resides in parts body, such oral cavity, nasal passages, lungs, gut, skin, bladder, vagina. gut gastrointestinal tract has received particular attention due to its high abundance role psychiatric neurodegenerative However, presents other tissues, though less abundant, also plays crucial immune system homeostasis, thus influencing development progression For example, imbalance associated periodontitis might increase risk for Additionally, studies using postmortem brain samples have detected widespread presence bacteria brains patients with Alzheimer’s disease. This article provides an overview emerging host disorders discusses future directions, underlying biological mechanisms, reliable biomarkers microbiota, microbiota-targeted interventions, this field.

To understand the dynamic interplay between human microbiome and host during health disease, we analyzed microbial composition, temporal dynamics, associations with multi-omics, immune, clinical markers of microbiomes from four body sites in 86 participants over 6 years. We found that stability individuality are body-site specific heavily influenced by host. The stool oral more stable than skin nasal microbiomes, possibly due to their interaction environment. identify individual-specific commonly shared bacterial taxa, individualized taxa showing greater stability. Interestingly, dynamics correlate across sites, suggesting systemic host-microbial-environment interactions. Notably, insulin-resistant individuals show altered among microbiome, molecular markers, features, disrupted metabolic disease. Our study offers comprehensive views multi-site relationship

Microneedles (MNs) can be used for the topical treatment of skin disorders as they directly deliver therapeutics to site lesions, resulting in increased therapeutic efficacy while having minimum side effects. MNs are different kinds (e.g., small molecules, macromolecules, nanomedicines, living cells, bacteria, and exosomes) treating various disorders, including superficial tumors, wounds, infections, inflammatory diseases, abnormal appearance. The improved by integrating advantages multiple perform combination therapy. Through careful designing, further modified with biomimetic structures responsive drug release from internal external stimuli enhance transdermal delivery efficiency robust outcomes. Some studies have proposed use drug-free a promising mechanotherapeutic strategy promote wound healing, scar removal, hair regeneration via mechanical communication pathway. Although several advantages, practical application suffers problems related industrial manufacture clinical evaluation, making it difficult translation.
