Gut microbiota and integrins are known to contribute colorectal cancer (CRC), but whether they interact has been unclear. Here, we provided evidence that Fusobacterium nucleatum upregulated integrin α5 (ITGA5) in CRC both human patients murine models. Abstract CD8 + T cells secreting granzyme A (GZMA) can induce pyroptosis in tumor by effectively cleaving gasdermin B (GSDMB), which is stimulated interferon-γ (IFN-γ). However, the interaction between GZMA-expressing and GSDMB-expressing colon cancer remains poorly understood. Our research employed multi-color immunohistochemistry (mIHC) staining integrated clinical data to explore spatial distribution relevance of GZMA- IFN-γ-expressing tumor-infiltrating lymphocytes (TILs), as well CK cells, within microenvironment (TME) human samples. Additionally, we utilizing single-cell RNA sequencing (scRNA-seq) examine functional dynamics interactions among these cell populations. scRNA-seq analysis colorectal (CRC) tissues revealed that TILs co-expressed GZMA IFN-γ, but not other types. mIHC results indicated a significant reduction infiltration IFN-γ patients ( P < 0.01). Functional demonstrated enhanced activation effector functions compared TIL subsets. Furthermore, exhibited augmented immunogenicity. Correlation highlighted positive association GSDMB r = 0.221, 0.033).
