LILRB4, a myeloid inhibitory receptor belonging to the family of leukocyte immunoglobulin-like receptors (LILRs/LIRs), plays pivotal role in regulation immune tolerance. LILRB4 primarily mediates suppressive responses by transmitting signals through immunoreceptor tyrosine-based motifs (ITIMs). This checkpoint molecule has gained considerable attention due its potent regulatory functions. Its ability induce effector T cell dysfunction and promote suppressor differentiation been demonstrated, indicating therapeutic potential for modulating excessive responses, particularly autoimmune diseases or induction transplant Additionally, intervening with molecules, system responsiveness can be adjusted, representing significant value areas such as cancer treatment. Thus, emerged key player addressing diseases, tolerance induction, other medical issues. In this review, we provide comprehensive overview encompassing structure, expression, ligand molecules well receptor. By exploring involvement various significance disease progression is emphasized. Furthermore, propose that manipulation represents promising immunotherapeutic strategy highlight prevention, treatment diagnosis.

Multiple sclerosis (MS) is an immune-inflammatory disease that attacks and damages myelinated axons in the central nervous system (CNS) causes nontraumatic neurological impairment young people. Historically, Lidwina of Schiedam documented first MS case. After that, Augustus d'Este wrote for years about how his symptoms worsened. Age, sex, genetics, environment, smoking, injuries, infections, including herpes simplex rabies, are risk factors MS. According to epidemiology, average age onset between 20 40 years. more prevalent women common Europe America. As diagnostic methods criteria change, people with may be discovered at earlier stages disease. therapy has advanced dramatically due breakthroughs our knowledge disease's etiology progression. Therefore, efficacy treatment medications increased exponentially. Management goals include reducing lesion activity avoiding secondary Current approaches focus on managing acute episodes, relieving symptoms, biological activity. Disease-modifying drugs such as fingolimod, interferon-beta, natalizumab, dimethyl fumarate most widely used treatments For proof safety these medications, investigations real world necessary.

Background There is increasing evidence that the types of immune cells are associated with various neurodegenerative diseases. However, it currently unclear whether these associations reflect causal relationships. Objective To elucidate relationship between and diseases, we conducted a two-sample Mendelian randomization (MR) analysis. Materials methods The exposure outcome GWAS data used in this study were obtained from an open-access database ( https://gwas.mrcieu.ac.uk/ ), employed MR analysis to assess 731 cell features four including Alzheimer’s disease (AD), Parkinson’s (PD), amyotrophic lateral sclerosis (ALS) multiple (MS). All was Multiple minimize bias obtain reliable estimates variables interest outcomes. Instrumental variable selection criteria restricted ensure accuracy effectiveness species risk Results identified potential relationships different Specifically, found 8 have AD, 1 type has PD, 6 ALS, MS. Conclusion Our study, through genetic means, demonstrates close specific ALS MS, providing useful guidance for future clinical researches.

Mitochondrial dysfunction is increasingly recognized as a key factor in neurodegenerative diseases (NDDs), underscoring the therapeutic potential of targeting mitochondria-related genes. This study aimed to identify novel biomarkers and drug targets for these through comprehensive analysis that integrated genome-wide Mendelian randomization (MR) with genes associated mitochondrial function. Using existing publicly available association studies (GWAS) summary statistics data on 1136 genes, we initially identified subset related function exhibited significant associations NDDs. We then conducted colocalization summary-data-based (SMR) analyses using expression quantitative trait loci (eQTL) validate causal role candidate Additionally, assessed druggability encoded proteins prioritize further exploration. Genetically predicted levels 10 were found be significantly risk Elevated DMPK LACTB2 increased Alzheimer's disease risk. Higher NDUFAF2, BCKDK, MALSU1, along lower TTC19, raised Parkinson's ACLY both amyotrophic lateral sclerosis multiple (MS) risks, while decreased MCL1, TOP3A, VWA8 MS These primarily impact energy metabolism. Notably, several druggable protein are being explored NDDs treatment. data-driven MR demonstrated this could serve pharmacological prevention treatment © 2025 International Parkinson Movement Disorder Society.

The pathophysiology of different neurodegenerative illnesses is significantly influenced by the polarization regulation microglia and macrophages. Traditional classifications macrophage phenotypes include pro-inflammatory M1 anti-inflammatory M2 phenotypes. Numerous studies demonstrated dynamic non-coding RNA modifications, which are catalyzed microglia-induced neuroinflammation. Different nutraceuticals focus on M1/M2 macrophages, offering a potent defense against neurodegeneration. Caeminaxin A, curcumin, aromatic-turmerone, myricetin, aurantiamide, 3,6'-disinapoylsucrose, resveratrol reduced microglial inflammatory markers while increased indicators in Alzheimer's disease. Amyloid beta-induced activation was suppressed andrographolide, sulforaphane, triptolide, xanthoceraside, piperlongumine, novel plant extracts also prevented microglia-mediated necroptosis apoptosis. Asarone, galangin, baicalein, a-mangostin oxidative stress cytokines, such as interleukin (IL)-1, IL-6, tumor necrosis factor-alpha M1-activated Parkinson's Additionally, myrcene, icariin, tenuigenin nod-like receptor family pyrin domain-containing 3 inflammasome neurotoxicity, a-cyperone, citronellol, nobiletin, taurine NADPH oxidase 2 nuclear factor kappa B activation. Furthermore, other like plantamajoside, swertiamarin, urolithin kurarinone, Daphne genkwa flower, Boswellia serrata showed promising neuroprotection treating In Huntington's disease, elderberry, iresine celosia, Schisandra chinensis, gintonin, pomiferin results improved patient symptoms. Meanwhile, linolenic acid, resveratrol, Huperzia serrata, baicalein protected activated macrophages experimental autoimmune encephalomyelitis multiple sclerosis. emodin, esters gallic rosmarinic acids, Agathisflavone, sinomenine offered sclerosis treatments. This review highlights therapeutic potential using to treat diseases involving microglial-related pathways.
