Abstract Inflammaging, characterized by an increase in low-grade chronic inflammation with age, is a hallmark of aging and strongly associated various age-related diseases, including liver disease (CLD) hepatocellular carcinoma (HCC). Because necroptosis cell death pathway that induces through the release DAMPs, we tested hypothesis age-associated contributes to liver. Phosphorylation MLKL MLKL-oligomers, markers necroptosis, as well phosphorylation RIPK3 RIPK1 were significantly upregulated livers old mice relative young this occurred later half life (i.e., after 18 months age). Markers M1 macrophages, expression proinflammatory cytokines (TNFα, IL6 IL-1β), fibrosis age change paralleled changes fibrosis. Hepatocytes macrophages isolated from showed elevated levels increased mice. Short term treatment inhibitor, necrostatin-1s (Nec-1s), reduced cytokines, Thus, our data show for first time liver, which turn appears contribute possibly CLD. Graphical abstract

The disparity between increased lifespan and healthy aging, marked by prevalent "inflammaging", highlights the global challenge in care of older persons. This study explored anti-inflammatory effects Lactiplantibacillus plantarum HEAL9 (LpHEAL9), alone or combined with berries, on volunteers chronic low-grade inflammation (LGI). It was a randomized, double-blind, placebo-controlled trial, total 66 (> 70 years old), randomly assigned, equally distributed, to placebo, LpHEAL9 + Berries group. After 2-week run-in period, participants underwent 4-week dietary intervention. Intake showed trend towards reduction serum CRP but without reaching statistical significance. However, significantly decreased fecal calprotectin levels compared placebo. LpHEAL9+Berries did not show any effect inflammation. Both probiotic groups improving cognitive function albeit Our findings suggest that strain L. has modest impact LGI population (ClinicalTrials.gov ID: NCT02342496).

Liver fibrosis might be linked to the prevalence of chronic kidney disease (CKD). However, there is little information about association between liver and decreased function in middle-aged older subjects. We aimed evaluate influence on incidence or CKD stage 3–5 a retrospective cross-sectional study (Study 1, n = 806) 6-year longitudinal 2, 380) evaluated using Fibrosis-4 (FIB-4) index estimated glomerular filtration rate (eGFR) all All subjects were divided into four groups basis their FIB-4 score quartiles (low high). In Jonckheere–Terpstra trend test Study eGFR significantly from lowest group highest (p < 0.001). The Kaplan–Meier survival curve 2 showed that cumulative was higher third quartile than other quartiles. Our results suggest could useful indicator for CKD, even within relatively healthy population, although not an independent risk factor.

Primary biliary cholangitis (PBC), and primary sclerosing (PSC) are rare immune-related cholangiopathies with still poorly explained pathogenesis. Although triggers of chronic inflammation subsequent fibrosis that affect cholangiocytes leading to obliteration bile ducts conversion liver cirrhosis unclear, both disorders regarded be multifactorial. Different factors can contribute the development hepatocellular injury in course progressive cholestasis, including (1) body accumulation acids their toxicity, (2) decreased food intake nutrient absorption, (3) gut microbiota transformation, (4) reorganized host metabolism. Growing evidence suggests intestinal microbiome composition not only altered by dysfunction, but turn, it actively impacts hepatic conditions. In this review, we highlight role key such as gut–liver axis, barrier integrity, acid synthesis circulation, composition, which seem strongly related PBC PSC outcome. Emerging treatments future therapeutic strategies also presented.

L-Citrulline (L-Cit) is discussed to possess a protective effect on intestinal barrier dysfunction but also diminish aging-associated degenerative processes. Here, the effects of L-Cit lifespan were assessed in C. elegans, while decline determined C57BL/6J mice. For analysis, elegans treated with ±5 mM L-Cit. Twelve-month-old male mice (n = 8–10/group) fed standard chow diet received drinking water ± 2.5 g/kg/d or 5 hydrolyzed soy protein (Iso-N-control) for 16 32 weeks. Additionally, 4-month-old accordingly 8 Markers senescence, glucose tolerance, function, and microbiota composition analyzed treatment significantly extended elegans. The significant increase markers senescence signs impaired tolerance found 16- 20-month-old control was attenuated L-Cit-fed mice, which associated protection from decrease NO2− levels small intestine, no marked differences when comparing age-matched groups. Our results suggest that pharmacological doses may have beneficial
